  1. 1. Altering HIV-1 envelope glycoprotein maturation and its effects on viral infectivity

    Författare :Alenka Jejcic; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :HIV-1;

    Sammanfattning : HIV-1 is dependent on its envelope glycoprotein (Env) to initiate infection. Env binds to cellular receptors and mediate the following fusion of the viral envelope with the cell plasma membrane. LÄS MER

  2. 2. Variable viral genes as genetic immunogens

    Författare :Karl Ljungberg; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Influenza; hemagglutinin; HIV-1; gp160; DNA vaccine; genetic immunization; CpG; GM-CSF; pseudovirus.;

    Sammanfattning : Influenza and HIV-1 are two highly variable viruses that cause extensive morbidity and mortality in many countries. The most cost-effective medical intervention against influenza is vaccination but this has to be repeated annually because of the low antigenicity of the vaccine. LÄS MER

  3. 3. Genetic vaccination against acute viral disease

    Författare :Marina N Fleeton; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Semliki Forest virus; Alphavirus. Influenza A virus; DNA vaccines. Influenza vaccine. Respiratory Syncytial Viruses; Louping Ill. Flavivirus Infections;

    Sammanfattning : This thesis describes the development of recombinant vaccines based on the Semliki Forest virus (SFV) expression system. Immunisation of mice with recombinant virus particles, a layered DNA/RNA plasmid vector, and recombinant self-replicating RNA were carried out and the protective effect of these recombinant vaccines against viral challenge were examined. 4. 4. Incorporation of cellular proteins into enveloped virus particles

    Författare :Maria Hammarstedt; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :MLV; HIV-1; assembly; protein sorting; HfIV-6A; autoimmunity; MS;

    Sammanfattning : This thesis work aimed to investigate the assembly and budding of enveloped virus particles with focus on the fate of cellular proteins, present in or near the plasma membrane (PM) where the budding occurs. It was previously shown that compact viruses, like alphaviruses, with a covering outer protein coat, did not contain any cellular proteins in the envelope. LÄS MER

  5. 5. Interaction-mediating sequences within class I viral fusion glycoproteins : their roles in viral infection and in applications

    Författare :Simin Zhang; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Class I viral fusion glycoproteins facilitate fusion of the viral envelope with cell membranes and entry of the virus into the cell, through extensive short sequence-specific interactions. Regions mediating these interactions include the N-terminal hydrophobic fusion peptide, a pair of extended 4,3-hydrophobic heptad repeats (HRs), a membrane-active membrane proximal external region (MPER), a hydrophobic transmembrane domain and the cytoplasmic tail region.
