  1. 6. Conditional and non-conditional reward-related responses to alcohol. Nicotinic mechanisms

    Författare :Elin Löf; Göteborgs universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; ethanol; nicotine; ventral tegmental area; nucleus accumbens; dopamine; nicotinic acetylcholine receptor; gamma-amino-butyric acid receptor A; conditioned reinforcement; in vivo microdialysis; rat;

    Sammanfattning : The mesolimbic dopamine system is believed to mediate the positive reinforcing and rewarding effects of addictive drugs by increasing dopamine levels in its terminal area, the nucleus accumbens (nAc). Nicotinic acetylcholine receptors (nAChRs) within this system appear important for the pharmacological actions of both alcohol and nicotine, which may explain the frequent co-abuse of these two drugs. 2. 7. Nicotinic effects on midbrain dopamine neurons : a dual mechanism of action

    Författare :Björn Schilström; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :nicotine; dopamine; ventral tegmental area; glutamate; microdialysis; Fos expression; NMDA receptor; presynaptic;

    Sammanfattning : The neurons of the mesocorticolimbic dopamine system are located in the ventral tegmental area (VTA) in the mesencephalon and send their projections to forebrain structures, such as the prefrontal cortex, the amygdala and the nucleus accumbens. The dopamine system is important in regulating mood and motivation as well as cognitive function and appear to be primarily involved in drug dependence, since most drugs abused by man stimulate this system. 3. 8. The phencyclidine model of schizophrenia : dysregulation of brain dopamine systems induced by NMDA receptor antagonists

    Författare :Jan M Mathé; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :phencyclidine; MK-801; CNQX; prazosin; tetrodotoxin; dopamine; neurotensin; ventral tegmental area; nucleus accumbens; medial prefrontal cortex;

    Sammanfattning : Non-competitive NMDA receptor antagonists, such as phencyclidine (PCP) and dizocilpine (MK-801) can even acutely induce a drug induced psychotic state which closely resembles schizophrenia. The midbrain dopamine (DA) neurons, which originate in the ventral tegmental area (VTA), are involved in motivational, attentional and cognitive processes, and have also been profoundly implicated in schizophrenia. 4. 9. Effects of nicotine on mesolimbocortical dopaminergic neurotransmission : a pharmacological study in the rat

    Författare :Magnus Nisell; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :nicotine; dopamine; ventral tegmental area; accumbens; prefrontal cortex; c-fos; drug-dependence; sensitization; mecamylamine; D; receptors; SCH 23390;

    Sammanfattning : Tobacco-smoking represents a form of drug addiction to nicotine (NlC) and the reinforcing and dependence-producing properties of NIC depend largely on central dopamine (DA) neurons, located in the ventral tegmental area (VTA) and projecting mainly to the nucleus accumbens (NAc; mesolimbic DA system) and the medial prefrontal cortex (PFC; mesocortical DA system). Generally, the mesolimbic DA system plays a pivotal role in the reinforcing effects of natural rewards as well as of drugs of abuse. LÄS MER

  5. 10. Neurobiological basis of the nicotine withdrawal reaction : an experimental analysis

    Författare :Bengt E Hildebrand; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :rat; nicotine; nicotine withdrawal; behavior; locomotor activity; microdialysis; intracerebral injections; mesolimbocortical; dopamine; ventral tegmental area; nucleus accumbens; amygdala; immediate-early genes; c-fos;

    Sammanfattning : The mesolimbocortical dopamine (DA) system is pivotal for the mediation of the reinforcing effects of many dependence-producing drugs. It consists of cell bodies in the ventral tegmental area (VTA) that project to e.g. the nucleus accumbens (NAC), the central nucleus of amygdala (CNA) and the medial prefrontal cortex. LÄS MER
