Sammanfattning : This thesis presents two new concepts in oral drug administration and the results of evaluation of some relevant formulation factors.Investigation into improving the homogeneity of mixtures for tableting indicated that it may be possible to obtain interactive dry mixtures of micronised drugs containing drug proportions as low as 0.015% w/w. 22. Pharmacodynamic modeling of cardiovascular effects of Σb-antagonists and moxonidine : Special emphasis on tolerance and rebound effects
Sammanfattning : Pharmacodynamic (PD) modeling was used to characterize and quantify the short- and long-term changes in cardiovascular effects following treatment with β-antagonists or imidazoline receptor agonist moxonidine, in spontaneous hypertensive rats and patients with congestive heart failure, respectively. The complex cardiovascular system is regulated by several feedback mechanisms with different time domains and gains. LÄS MER

Sammanfattning : The nasal administration of drugs offers advantages over administration by intravenous injection. Drugs can be rapidly absorbed through the nasal mucosa, resulting in a rapid onset of action, and also avoiding degradation in the gastrointestinal tract and first-pass metabolism in the liver. 24. The pharmacokinetics and pharmacodynamics of clevidipine, a new ultrashort-acting calcium antagonist, in different animal species and man
Sammanfattning : Clevidipine is a new ultrashort-acting calcium antagonist of the dihydropyridine type developed for reduction and control of blood pressure in connection with cardiac surgery. The aim of this thesis was to characterise the in vitro hydrolysis rate of clevidipine in different biological matrices and the pharmacokinetics and pharmacodynamics in animal species and various human populations.
