11. Computational models for the prediction of intestinal membrane permeability
Sammanfattning : Lead compounds generated in high-throughput drug discovery programs often have unfavorable biopharmaceutical properties, resulting in a low success rate for such drug candidates in clinical development. Efficient and reliable methods that predict biopharmaceutical properties, such as intestinal permeability and solubility are therefore required in order to reduce the attrition rate during development of these compounds. 12. Preparation and characterisation of biodegradable microspheres for the delivery of bioactive compounds
Sammanfattning : The incorporation of bioactive compounds into biodegradable polymer microspheres has several advantages. Firstly, the drug substance is protected from physiological degradation. Secondly, using microspheres from polymers with different degradation mechanisms, it is possible to design different release profiles with desired release rates. LÄS MER
13. Nasal Drug Delivery : In Vitro Studies on Factors Influencing Permeability and Implications on Absorption
Sammanfattning : Nasal delivery is a feasible alternative to oral or parenteral administration for some drugs because of the high permeability of the nasal epithelium, rapid drug absorption across this membrane and avoidance of hepatic first-pass metabolism. The main objective of this thesis was to investigate factors influencing the permeability of the nasal mucosa to various compounds and to evaluate implications for drug absorption via the nasal route. 14. Formulations, Release and Skin Penetration of Topical Anesthetics
Sammanfattning : This thesis describes certain critical aspects of the development of semisolid topical anesthetic formulations requiring a fast onset of action. Furthermore, local anesthetics were investigated regarding their phase interaction with membrane lipids. LÄS MER
15. The mucus layer and other barriers to intestinal drug absorption
Sammanfattning : The extraepithelial mucus layer acts as a barrier to the absorption and diffusion of drugs. Three in vitro models have been used for studies of this and other intestinal barriers to drug transport. A drug absorption model based on monolayers of the mucus-producing human intestinal goblet HT29-H cell line was developed. LÄS MER
