1. Autoantibodies and the Type I Interferon System in the Etiopathogenesis of Systemic Lupus Erythematosus
Sammanfattning : In sera remitted for anti-nuclear antibody (ANA) analysis, the supplement of a sensitive anti-SSA/Ro ELISA to the conventional ANA screening by immunofluorescence (IF) revealed that one fourth of the individuals with IF-ANA negative, but SSA/Ro ELISA positive sera, had systemic lupus erythematosus (SLE) or cutaneous LE. Consequently, adding a sensitive anti-SSA/Ro ELISA to the ANA screening is valuable for the serological detection of ANA negative SLE/LE patients. LÄS MER
2. Endogenous Type I Interferon Inducers in Systemic Autoimmune Diseases
Sammanfattning : Patients with systemic lupus erythematosus (SLE) have elevated levels of interferon (IFN)-α in blood and IFN-α-producing cells in tissues. In the present thesis, we investigate the mechanisms behind the upregulated IFN-α-production in SLE and also show that the IFN-α system is activated in primary Sjögren’s syndrome (pSS), with IFN-α-producing cells in the major affected organ, the salivary glands. 3. Large-Scale Genotyping for Analysis of the Type I Interferon System in Autoimmune Diseases
Sammanfattning : Single nucleotide polymorphisms (SNPs) are the most common form of genetic variation. We developed a novel multiplexed method for SNP genotyping based on four-color fluorophore tag-microarray minisequencing. This method allows simultaneous genotyping of 80 samples and up to 200 SNPs in any allele combination. LÄS MER
4. Induction and functional role of type I interferon in bacterial infection of macrophages
Sammanfattning : Infection represents an evolutionary arms race between pathogen and host. Thus, scientists find that pathogenic microbes have evolved elegant strategies to manipulate or avoid our immune system, allowing them to persist within the human population. LÄS MER
5. Autoantibodies and the type I interferon system in idiopathic inflammatory myopathies
Sammanfattning : Idiopathic inflammatory myopathies (IIM), also known as myositis, are rare autoimmune diseases, characterized by proximal muscle weakness and inflammatory cells in skeletal muscle tissue. The most common subgroups are polymyositis (PM), dermatomyositis (DM) and inclusion body myositis (IBM). LÄS MER
