  1. 1. Mating type switching and transcriptional silencing in Kluyveromyces lactis

    Författare :Emad Barsoum; Åström Stefan; Laura Rusche; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Mating type switch; transcriptional silencing; alpha3; transposase; Ume6; Mts1; Ras; cAMP; K. lactis; Developmental biology; Utvecklingsbiologi; utvecklingsbiologi; Developmental Biology;

    Sammanfattning : To explore the similarities and differences of regulatory circuits among budding yeasts, we characterized the role of unscheduled meiotic gene expression 6 (UME6) and a novel mating type switching pathway in Kluyveromyces lactis. We found that Ume6 was required for transcriptional silencing of the cryptic mating-type loci HMLα and HMRa. LÄS MER

  2. 2. Silencing and DNA double-strand break repair in budding yeast

    Författare :Andreas Kegel; Stefan Åström; Stephan Marcand; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Silencing; Reb1; DSB; NHEJ; Nej1; illegitimate recombination; yeast; Developmental biology; Utvecklingsbiologi;

    Sammanfattning : Transcriptional silencing that makes large chromosomal domains inaccessible for the transcriptional apparatus is nucleated at DNA elements called silencers. In K.lactis a 102bp HMLα silencer was defined revealing three distinct protein-binding regions (A, B, and C) that were required for silencing of HMLα. LÄS MER

  3. 3. Mechanistic characterization of post-transcriptional gene silencing

    Författare :Cecilia Dahlgren; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : With the ongoing task of identifying and characterizing genes in the human genome, there is a great demand for tools to study these genes. Today, reverse genetics is the most effective way to explore gene function. This involves suppression of the gene of interest by a set of manipulation techniques. LÄS MER

  4. 4. On aging and the role of Ubp3 in heterochromatic gene silencing and protein quality control

    Författare :David Öling; Göteborgs universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Aging; Protein damage; Segregation; Transcriptional silencing; rDNA; UBP3; SIR2; Protein aggregates;

    Sammanfattning : Aging is characterized by a build-up of damage in organisms ranging from protists to multi-cellular species. This damage adversely affects core components such as DNA and proteins which are necessary to sustain life. LÄS MER

  5. 5. Molecular Insights into Kcnq1ot1 Noncoding Antisense RNA Mediated Long Range Transcriptional Gene Silencing

    Författare :Radha Raman Pandey; Chandrasekhar Kanduri; Karl Ekwall; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; epigenetics; gene silencing; noncoding RNA; genomic imprinting; Kcnq1ot1; Medical genetics; Medicinsk genetik;

    Sammanfattning : Non-coding antisense RNAs have been implicated in the epigenetic silencing of individual gene as well as chromosomal domains. While silencing of the overlapping gene by antisense RNAs has been well investigated, their functional role in silencing of chromosomal domains remains enigmatic. LÄS MER
