1. Transcriptional and epigenetic control of gene expression in embryo development
Sammanfattning : During cell specification, temporal and spatially restricted gene expression programs are set up, forming different cell types and ultimately a multicellular organism. In this thesis, we have studied the molecular mechanisms by which sequence specific transcription factors and coactivators regulate RNA polymerase II (Pol II) transcription to establish specific gene expression programs and what epigenetic patterns that follows. 2. Regulation of gene expression in fruit flies : how does it start, and will it be remembered?
Sammanfattning : One of the most distinctive features of eukaryotic chromosomes is the bundling of DNA together with functionally associated RNA and proteins in chromatin. This allows huge amounts of DNA to be packed inside the very tiny space of the nucleus, and alterations in the structure of chromatin enable access to the DNA for transcription (“reading” genes by production of RNA copies). 3. The interplay of transcription and chromatin regulation during embryonic development
Sammanfattning : A fascinating aspect of multicellular development is the production of diverse cell types from an identical DNA genome, which depends on the ability to express different complements of genes across space and time. This thesis tries to address how the genome, within the chromatin structure that envelops it, is regulated to direct this complex process. LÄS MER
4. The odyssey of MYC transcript from the nucleus to the cytoplasm : the molecular mechanism of the gene gating in human cancer cells
Sammanfattning : Pathological expression of the MYC oncogene is a common denominator in a wide range of cancers and is linked with abnormal cell proliferation. To achieve this status, the MYC gene benefits from being embedded in a region rich in enhancers and super-enhancers that are often absent in the normal cell counterparts. LÄS MER
5. The Role of the Escherichia coli RNA Polymerase β´Jaw in Transcription
Sammanfattning : Transcription, the central step in gene expression and regulation, is carried out by the DNA-dependent RNA polymerase (RNAP). Bacterial RNAP is a complex molecular machine in which the network of interacting parts and their movements, including contacts to nascent RNA and the DNA template, are at best partially understood.
