  1. 1. Activation and Regulation of TRPV1 : Studies in Recombinant Human Neuroblastoma TRPV1-SHSY5Y Cells

    Författare :Johanna EL Andaloussi-Lilja; Anna Forsby; Antonio Ferrer-Montiel; Stockholms universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; nociception; neurotrophic factors; eye irritation; stable transfection; Ca2 ; Neurochemistry; Neurokemi; Neurochemistry and Molecular Neurobiology; neurokemi med molekylär neurobiologi;

    Sammanfattning : TRPV1 is a transmembrane non-selective cation channel with preference for Ca2+. The receptor is primarily localised on dorsal root ganglion neurons and is activated by numerous endogenous and exogenous potentially irritating ligands eliciting pain. LÄS MER

  2. 2. Sensory mechanisms in the human upper airway - Role of TRP ion channels

    Författare :Lisa Alenmyr; Avdelningen för klinisk kemi och farmakologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; TRPV1; TRPV4; TRPA1; allergic rhinitis; itch; rhinorrhea; MUC5B; MUC5AC; epithelial;

    Sammanfattning : The nasal mucosa is richly innervated with sensory nerves having the ability to detect changes in the environment and mediate symptoms such as secretion and itch. Some TRP ion channels such as TRPV1 and TRPA1 are abundantly expressed in sensory nerves and respond to a wide variety of stimuli, including irritants, inflammatory mediators and temperatures. 3. 3. Bioactive Lipids in Nociception

    Författare :Anna Ermund; Avdelningen för klinisk kemi och farmakologi; []
    Nyckelord :Cannabinoids; Anandamide; AM404; 2-Arachidonoylglycerol; Acetaminophen; Cyclooxygenase; Nocicieption; TRPV1;

    Sammanfattning : This thesis focuses on bioactive lipids as (1) metabolites of the widely used antipyretic and analgesic drug acetaminophen and (2) activators of the ion channel TRPV1, an important downstream target for inflammatory mediators, in the phospholipase C (PLC)/TRPV1 signaling pathway. Evidence is presented for a fatty acid amide hydrolase (FAAH)-dependent fatty acid conjugation of p-aminophenol, a known acetaminophen metabolite, to form the potent TRPV1 activator AM404 in the central nervous system. 4. 4. Methods for elucidating membrane protein structure and function

    Författare :Carolina Trkulja; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; patch clamp; antibodies; mass spectrometry; ion channels; proteomics; fluorescence; TRPV1; cholesterol.; limited proteolysis; Membrane proteins;

    Sammanfattning : Elucidation of the structure-function relationship of membrane proteins increases our understanding of many biological processes, both physiological and pathological. However, membrane proteins have been notoriously difficult to study, having part of their structure embedded into the lipid cell membrane. LÄS MER

  5. 5. On methods for probing structure and function of membrane proteins

    Författare :Erik Jansson; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; proteomics; patch-clamp; mass spectrometry.; proteoliposomes; microfluidic superfusion; cholesterol; TRPV1; limited proteolysis; Membrane proteins; fluorescence;

    Sammanfattning : The determination of structure–function relationships in membrane proteins plays a key role for the understanding of molecular mechanisms in biological systems, and may provide information on how these systems can be modulated by pharmaceuticals. The aim of the work described herein was to develop methods for studying structure and function of membrane proteins, ideally in combination. LÄS MER
