    Sammanfattning : In this thesis methods for investigation of orientation and conformation of individual macromolecules on surfaces are presented as well as novel methods for functionalization of silicon chips with the possibility to get an ordered immobilization of antibodies.     Sammanfattning : Confocal microscopy has been used as a tool for studying adsorption of biomolecules to individual chromatographic adsorbent particles.     Sammanfattning : The work described in this thesis aims to achieving surface patterning through chemical activation of thiolated silicon oxide surfaces, resulting in a spatially controlled covalent immobilization of biomolecules with high resolution.Existing chemical methods to immobilize molecules on surfaces do not reach below the micrometer scale while the ones allowing for spatial control mostly lead to non-covalent adsorption of molecules on surfaces, or require several successive chemical reactions to obtain the final covalent immobilization.     Sammanfattning : Two polysaccharide-degrading enzymes (endo-1,4-D-glucanase and β-mannanase) from blue mussel, Mytilus edulis, have been purified to homogeneity using a combination of several chromatographic steps. Each enzyme has been characterized with regard to its molecular weight, isoelectric point, pH and temperature stability, pH and temperature optimum and substrate specificity. LÄS MER
