11. Microbial adaptations and controlling mechanisms of surface-associated microhabitat heterogeneity in aquatic systems
Sammanfattning : Habitat heterogeneity is a driving factor for speciation and ecosystem functioning and is well studied in macro-ecology. Yet our understanding of microbial adaptations, and governing processes is incomplete. LÄS MER
12. Thermodynamics of protein-DNA interactions
Sammanfattning : This thesis treats the molecular recognition between DNA-binding proteins and DNA and the way these interactions are affected by the surrounding water solution and dissolved solutes. The work involves studies on the thermodynamics of sequence specific DNA-binding of the human glucocorticoid receptor DNA-binding domain and the nonsequence specific DNA- binding of Sso7d from the archaeon Sulfolobus solfataricus. 13. Shining light on unknown function proteins
Sammanfattning : The study of protein structure and function is a fundamental step for understanding biological systems and for creating a solid ground for future drug development. Despite the development of novel ground-breaking biochemical and bioinformatical tools in the last decades, most proteins found in nature still have unknown assigned function. LÄS MER
14. Beta-Glycosidases from Hyperthermophiles as Biocatalysts
Sammanfattning : The thesis concerns glycosyl transfer with use of b-glycosidases, most of them from hyperthermophilic organisms. Both optimisation of the reaction conditions and protein engineering in terms of site-directed mutagenesis were used to improve b-glycosidase-catalysed reactions. LÄS MER
15. Functional genomics at the interface of protein expression and biophysical analysis
Sammanfattning : Protein characterization is central to any functional genomics effort, hence protein expression and purification play fundamental roles in these initiatives. In this study strategies for protein expression in E. coli were analysed, and several parameters were investigated respect to high-level expression of functional proteins. LÄS MER
