  1. 1. Covariate Model Building in Nonlinear Mixed Effects Models

    Författare :Jakob Ribbing; E. Niclas Johnsson; Mats O. Karlsson; Janet Wade; Uppsala universitet; []
    Nyckelord :Pharmacokinetics Pharmacotherapy; Pharmacokinetics; Pharmacodynamics; Modeling; Covariate selection; Stepwise selection; Covariate analysis; Methodology; Model validation; Model evaluation; Type-2 diabetes; Beta-cell function; Meta analysis; Cross-validation; Least absolute shrinkage and selection operator; Pharmacometrics; ED optimization; Farmakokinetik Farmakoterapi;

    Sammanfattning : Population pharmacokinetic-pharmacodynamic (PK-PD) models can be fitted using nonlinear mixed effects modelling (NONMEM). This is an efficient way of learning about drugs and diseases from data collected in clinical trials. LÄS MER

  2. 2. Molecular and Genetic Evidence for Antigen Selection in the Pathogenesis of Chronic Lymphocytic Leukemia

    Författare :Lesley Ann Sutton; Richard Rosenquist; John Gribben; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; IGHV4-34; microenvironment; stereotypy; immunoglobulin; intraclonal diversification; clonal evolution;

    Sammanfattning : Antigens play a critical role in the development of chronic lymphocytic leukemia (CLL) by binding to and stimulating leukemic precursor cells at some point during CLL ontogeny. Nevertheless, much remains unknown and further studies are necessary before an accurate model of antigen-drive can be ascertained. LÄS MER

  3. 3. The role of cortico-basal ganglia systems in the construction of movement: Action selection and sequencing

    Författare :Joel Sjöbom; Integrativ neurofysiologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Neurophysiology; Electrophysiology; Voluntary movements; In-vivo; Motor Cortex; Corticostriatal; Basal Ganglia;

    Sammanfattning : The cortico-basal ganglia system is instrumental in the construction of movement, through its involvement in aspects such as action selection, initiation/termination and sequencing, though the precise role of the different structures and how they collaborate on a network level to allow us to fluidly change from one behavior to another has long proven to be elusive.We have here recorded neuronal activity throughout the cortico-basal ganglia circuit in rats, in a series of projects in order to understand more about how some of these functions are controlled. 4. 4. Identification, validation and clinical application of a three-gene signature for accurate prognosis prediction and treatment selection of newly diagnosed prostate cancer

    Författare :Zhuochun Peng; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : This thesis presents a complete, comprehensive and stepwise approach of translational molecular research. Starting from whole-genome bioinformatics analyses based on the embryonic stem (ES) cell hypothesis, a three-gene signature was identified, validated with the goal of clinical application in order to optimize treatment decision based on improvement of overall survival estimation. 5. 5. Methodological Studies on Covariate Model Building in Population Pharmacokinetic-Pharmacodynamic Analysis

    Författare :Ulrika Wählby; Thomas Ludden; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical biosciences; Farmaceutisk biovetenskap; Biopharmacy; Biofarmaci; farmakokinetik och läkemedelsterapi; Pharmacokinetics and Drug Therapy;

    Sammanfattning : Population pharmacokinetic (PK) – pharmacodynamic (PD) modelling, using nonlinear mixed effects models, is increasingly being applied to obtain PK-PD information in drug development. Covariate modelling, the establishment of relationships between model parameters and patient characteristics, is undertaken to explain PK-PD variability and facilitate dose adjustment decisions, and is consequently an important objective of population PK-PD.
