1. Novel planar and particle-based microarrays for point-of-care diagnostics
Sammanfattning : Point-of-care assays are easy-to-use, portable and inexpensive tests that canbe used to aid diagnostics by measuring levels of disease-specific moleculesin settings where access to advanced laboratory equipment and trainedpersonnel are limited, such as at the patient's bedside or in low resourceparts of developing countries. In order to achieve high multiplexingcapacities, such assays can be based on planar microarrays consisting ofspots immobilized on a flat surface or on particle-based microarrays basedon populations of encoded particles. 2. Improved Techniques for High-Throughput Molecular Diagnostics
Sammanfattning : The amount of information derived from sequencing the humangenome is leading to an exponential increase in the rate atwhich genes and genetic disorders are mapped and characterized. As a consequence, the demand for genetic testing is alsodramatically increasing. LÄS MER
3. Molecular Tools for Biomarker Detection
Sammanfattning : The advance of biological research promotes the emerging of new methods and solutions to answer the biological questions. This thesis describes several new molecular tools and their applications for the detection of genomic and proteomic information with extremely high sensitivity and specificity or simplify such detection procedures without compromising the performance. 4. Molecular diagnostics of infectious diseases
Sammanfattning : In DNA-based diagnostics, the polymerase chain reaction(PCR) is the most widely used DNA amplification method. Toenable both sensitive and specific detection of agents causinginfectious diseases, the PCR needs to becombined with methodsto prepare the clinical sample containing the genetic materialof the pathogen. 5. Retroviral and related nucleic acid processing enzymes expression and detection : Applications on clinical studies of HIV and HERVs
Sammanfattning : Reverse transcriptase (RT), the expression and detection of the enzyme marking the family Retroviridae, has been studied. A new RT activity assay adapted to 96-well microtitre plates is described. It uses a non- radioactive bromodeoxyuridine triphosphate (BrdUTP) substrate in a solid phase format. The assay was highly sensitive. LÄS MER
