    Sammanfattning : Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a multipotent cytokine involved in the production and function of hematopoietic cells, and GM-CSF plays in particular a major role in responses to infection and physiological and pathological inflammatory processes. GM-CSF is produced in many cell types, and increases in the intracellular Ca2+ concentration are, like in many other systems, of major importance in the intracellular signaling that determines GM-CSF expression after receptor stimulation of the cells.     Sammanfattning : Transforming Growth Factor Beta (TGF-β) is a cytokine regulating a wide range of cellular processes such as proliferation, differentiation, and migration. At the early stages of cancer development TGF-β functions as a tumor suppressor, mainly due to its inhibitory effect on cellular growth, but during cancer progression, mutations in TGF-β signal components switches TGF-β into a promoter of cancer cell proliferation, survival and metastasis.     Sammanfattning : Hepatocellular carcinoma (HCC) is one of the most prevalent cancer types in the Western world and in the Asia-pacific regions, with its incidence expected to rise up to 22 million cases till 2020. Hepatocellular carcinoma etiology is mainly due to hepatitis B (HBV) and hepatitis C (HCV) infections, and to a lesser extent it is determined by the development of alcohol-driven cirrhosis and non-alcoholic steatohepatitis (NASH).     Sammanfattning : In the present thesis, genetic abnormalities in pancreatic cancer were studied, with special emphasis on alterations of components involved in the transforming growth factor beta (TGFB) signaling pathway. In the first study, fluorescence in situ hybridization and cytogenetic analyses revealed aberrations of chromosome 18 in all 13 pancreatic carcinoma cell lines studied, in particular frequent breaks close to the centromere of chromosome arm 18q.     Sammanfattning : Keloids are a fibroproliferative disorder of unknown etiology developing in the skin after injury or spontaneously. The aim of this thesis is to gain deeper insight into the role of TGF-β and its signaling pathway proteins, SMADs, in the pathogenesis of keloids and describe the gene expression profile in different keloid sites in the search for potential target genes for future treatment.
