  1. 6. Towards Gene Therapy of Osteopetrosis

    Författare :Maria Askmyr; Avdelningen för molekylärmedicin och genterapi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; extracellular fluids; Haematology; oc oc mice; osteoclast; hematopoietic stem cells; Hematologi; extracellulära vätskor;

    Sammanfattning : The goal in this thesis is development of gene therapy for malignant infantile osteopetrosis (IMO), a rare but severe genetic bone disease. The concept of osteopetrosis implies dysfunction or lack of osteoclasts, the bone resorbing cells in our body, resulting in failure of normal bone breakdown. LÄS MER

  2. 7. Development of Gene Therapy for Hematopoietic Stem Cells using Lentiviral Vectors

    Författare :Niels-Bjarne Woods; Andreas Ooka; Stefan Karlsson; Avdelningen för molekylärmedicin och genterapi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; embryonic stem cell; Hematopoietic stem cell; lentiviral vector; transduction efficiency; transgene expression; multiple vector copy integration; insertional mutagenesis; NOD SCID mouse; cytogenetics; Genetics; extracellulära vätskor; extracellular fluids; cytogenetik; Genetik; Haematology; Hematologi; Lentivirus : genetics; Human; Genetic Vectors; Hematopoietic Stem Cells : metabolism; Animal; Gene Therapy; Leukemia : therapy; Leukemia : metabolism;

    Sammanfattning : Hematopoietic stem cells are an ideal target for genetic manipulation for the purpose of curing hematological disorders as they have the ability to develop into all blood lineages and to self-renew. In this study we demonstrate that lentiviral vectors, based on HIV-1, can efficiently transfer genes into human hematopoietic progenitor and stem cells as assessed following stem cell transplantation in immune compromised mice. 3. 8. Retograde migration of starch particles in the female genital tract

    Författare :Anette Sjösten; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Gloves; glove-powder; starch particles; post-operative adhesions; vaginal examination; endometriosis; cell-culture; SCID-mice.;

    Sammanfattning : Tubal and pelvic adhesions are a major cause of female infertility in humans. The majority of postoperative intra-abdominal adhesions result from the presence of ischaemic tissue within the abdominal cavity. These precipitate an inflammatory vascular response from neighbouring healthy tissue. LÄS MER

  4. 9. Persistence of polyomavirus : versus immunity to polyoma tumors

    Författare :Zsofia Berke; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :polyomavirus persistence; CD4--; CD8--; IgM--; XID; SCID; TSTA; T-antigen; tumor rejection.;

    Sammanfattning : The aim of this thesis was to understand the role of different specific immune effectors with regard to polyomavirus persistence and polyomavirus induced tumor development. Furthermore, an MHC class I restricted short viral peptide was identified as a target for polyomavirus specific recognition of polyoma tumor cells. LÄS MER

  5. 10. The therapeutic potential of ex vivo expanded natural killer (NK) cells for immunotherapy of cancer

    Författare :Hayrettin Guven; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Natural killer NK cells; Natural killer-like T NKT cells; Gene transfer; Retrovirus; Good manufacturing practice GMP ; Ex vivo; In vivo; SCID-beige.;

    Sammanfattning : Cell and gene therapy of cancers has received much attention in the past decade. A number of applications of cancer treatment have been developed and used. One of the recent applications is immunotherapy of tumours with human natural killer (NK) cells. LÄS MER
