  1. 1. The hematopoietic transcription factor RUNX1 : a structural view

    Författare :Stefan Bäckström; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Molecular medicine; RUNX1; Runt domain; CBFβ; transcription factor; leukaemia; protein crystallography; anomalous diffraction; Molekylärmedicin; Molecular medicine genetics and pathology ; Molekylär medicin genetik och patologi ; molekylärbiologi; Molecular Biology;

    Sammanfattning : The malfunction of the transcriptional regulator RUNX1 is the major cause of several variants of acute human leukemias and its normal function is to regulate the development of the blood system in concert with other transcriptional co-regulators. RUNX1 belongs to a conserved family of heterodimeric transcription factors that share a conserved DNA binding domain, the Runt domain (RD), named after the first member of this group – Runt - found in Drosophila melanogaster. 2. 2. Structure-function relationships of the human Runx1 transcription factor

    Författare :Magnus Wolf-Watz; KTH; []
    Nyckelord :Runx1; Runt domain; CBFâ; transcription; leukemia; protein structure; protein dynamics;

    Sammanfattning : Runx transcription factors contain a DNA-binding andtransactivating α-subunit and a β-subunit (CBFβ)that serves to increase the DNA binding affinity of theα-subunit. The DNA binding domain of Runx proteins isnamed the Runt homology domain (RD) after its strong sequencehomology to theDrosophilaprotein Runt. LÄS MER

  3. 3. RUNX1/AML1 functions and mechanisms regulating granulocyte-macrophage colony-stimulating factor transcription

    Författare :Hebin Liu; Sven Påhlman; Umeå universitet; []
    Nyckelord :RUNX1 AML1; Granulocyte-macrophage colony-stimulating factor GM-CSF ; Calcineurin; CaMKII; Ets1; Transforming growth factor-b TGF-b ; Smad3; molekylärbiologi; Molecular Biology;

    Sammanfattning : Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a multipotent cytokine involved in the production and function of hematopoietic cells, and GM-CSF plays in particular a major role in responses to infection and physiological and pathological inflammatory processes. GM-CSF is produced in many cell types, and increases in the intracellular Ca2+ concentration are, like in many other systems, of major importance in the intracellular signaling that determines GM-CSF expression after receptor stimulation of the cells. 4. 4. Genomic characterization of ETV6/RUNX1-positive acute lymphoblastic leukemia

    Författare :Henrik Lilljebjörn; Avdelningen för klinisk genetik; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; SNP array; array CGH; ETV6 RUNX1-positive ALL; childhood ALL; exome sequencing;

    Sammanfattning : The t(12;21) translocation generates the ETV6/RUNX1 fusion gene, present in 25% of childhood acute lymphoblastic leukemia. This fusion gene is important for leukemia development but is not sufficient for leukemia to arise. LÄS MER

  5. 5. Stem and progenitor cell involvement in acute lymphoblastic leukemia

    Författare :Anders Castor; Stamcellscentrum (SCC); []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; cancer; Haematology; Cytology; oncology; cancerology; Cytologi; onkologi; Medicin människa och djur ; Medicine human and vertebrates ; ETV6-RUNX1; BCR-ABL1; stem cells; Acute lymphoblastic leukemia; extracellular fluids; Hematologi; extracellulära vätskor;

    Sammanfattning : Leukemic stem cells (LSCs) share the capacity of self renewal and extensive proliferation with normal hematopoietic stem cells (HSCs), and are therefore obvious targets for therapy. As such, they need to be identified and characterized in order to elucidate what drives them, and what separates them from their normal counterparts. LÄS MER
