    Författare :Oommen P. Varghese; Lars Engman; Jacek Stawinski; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Organic chemistry; chemically modified oligonucleotides; azetidine; aza-ENA-T; cyanomethylene locked; free energy of activation; diaxial interactions; chair conformation; stable duplex; human serum; snake venom phosphodiesterase; antisense agents; RNase H; Organisk kemi; Chemistry; Kemi;

    Sammanfattning : This thesis is concerned with synthesis, structure and biochemical analysis of chemically modified oligonucleotides with potential therapeutic applications. The three types of chemical modifications described here are: (a) A North-East locked 1',2'-azetidine nucleoside (b) A North locked 2',4'-cyanomethylene bridged nucleoside and (c) A 2',4'-aza-ENA-T nucleoside. 2. 7. Conformationally Constrained Oligonucleotides for RNA Targeting

    Författare :Qing Li; Jyoti Chattopadhyaya; Li-He Zhang; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; conformationally constrained nucleoside; antisense oligonucleotide; RNA affinity; nuclease stability; RNase H; RNA degradation; Chemistry with specialization in Bioorganic Chemistry; Kemi med inriktning mot bioorganisk kemi;

    Sammanfattning : A short oligonucleotide sequence as in a single-stranded antisense oligo nucleotides (AON) or in double-stranded small interfering RNAs (siRNA) can modulate the gene expression by targeting against the cellular mRNA, which can be potentially exploited for therapeutic purposes in the treatment of different diseases. In order to improve the efficacy of oligonucleotide-based drugs, the problem of target affinity, nuclease stability and delivery needs to be addressed. LÄS MER

  3. 8. Aspects of Antisense and Antigene Chemistry of Oligonucleotides Tethered to Intercalators

    Författare :Dimitri Ossipov; Jesper Wengel; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Bioorganic chemistry; oligonucleotides; intercalators; ruthenium complexes; RNase H cleavage; photocross-linkage; Bioorganisk kemi; Bioorganic chemistry; Bioorganisk kemi; bioorganisk kemi; Biorganic Chemistry;

    Sammanfattning : Synthetic and physicochemical studies on appropriately functionalized ODN-conjugates have been performed to evaluate their abilities to act as antisense agents against RNA or as intramolecular DNA cross-linking agents. Intercalating aromatic systems [phenazine (Pnz), dipyridophenazine (DPPZ)] and metallointercalators such as Ru2+(phen)2(DPPZ) and Ru2+(tpy)(DPPZ)L [where L = chemically or photochemically labile ligand, phen = phenanthroline, tpy = terpyridine], which are covalently tethered to the oligo-deoxynucleotides (ODNs), have been chosen for this purpose. 4. 9. Molecular insights into primer removal during mtDNA replication

    Författare :Ali Al-Behadili; Göteborgs universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Mitochondria; Mitochondrial DNA replication; DNA replication; Primer removal; RNase H1; Primer formation; Replication initiation; mitochondrial diseases;

    Sammanfattning : Mitochondria are vital for cell survival, and the primary producers of ATP, the energy currency used for various metabolic processes. Mitochondria are unique from other cellular compartments because they have their own genomes of circular small double-stranded DNA (mtDNA) of approximately 16. LÄS MER

  5. 10. Some Aspects of Nucleic Acids Chemistry

    Författare :Edouard Zamaratski; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Bioorganic chemistry; Bioorganisk kemi; Bioorganic chemistry; Bioorganisk kemi; bioorganisk kemi; Biorganic Chemistry;

    Sammanfattning : This thesis is divided into two parts based on a total of 8 papers: Part 1: Synthesis, physicochemical and biochemical studies of chemically modified oligonucleotides and their duplexes and triplexes. Potency of the chromophore conjugated DNA oligonucleotides as antigene and antisense gene repressors was evaluated. LÄS MER
