  1. 1. Versatile and Antique World of RNA : The Simplicity of RNA Mediated Catalysis

    Författare :Ema Kikovska; Leif Kirsebom; Venkat Gopalan; Uppsala universitet; []
    Nyckelord :Microbiology; RNA Catalysis; RNase P; Metal ions; tRNA Processing; Ribozyme; Mikrobiologi;

    Sammanfattning : RNA is the only biological molecule that can function both as a repository of information and as a catalyst. This, together with the ability to self-replicate, led to recognition of RNA as ‘prelude to life’.My work highlights some of the important features of RNA as a catalyst, exemplified by RNase P. 2. 2. iLocks: a novel tool for RNA assays with improved specificity

    Författare :Tomasz Krzywkowski; Mats Nilsson; Gunnar von Heijne; Weng-Onn Lui; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; RNA; miRNA; non-coding RNA; padlock probes; rolling circle amplification; invader; single cell; in situ; adenovirus; virology; diagnostics; Biochemistry; biokemi;

    Sammanfattning : The Central Dogma of molecular biology describes a framework for how genetic information is transferred in cells, placing RNA as a messenger between DNA and translated proteins. During the last years, interest in RNA research has grown tremendously due to the increasing understanding and recognition of the importance of RNA in regulation of gene expression, biochemical catalysis, and genome integrity surveillance. 3. 3. Distal to Proximal—Functional Coupling in RNase P RNA-mediated Catalysis

    Författare :Shiying Wu; Leif A. Kirsebom; Britt-Marie Sjöber; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Ribozyme; RNase P; Induced fit model; tRNA progressing; Substrate interaction;

    Sammanfattning : RNase P is a ubiquitous ribonuclease responsible for removing the 5’ leader of tRNA precursor. Bacterial RNase P contains one RNA (RPR) and one protein (RPP) subunit. However, the number of protein variants depends on the origin. The RNA subunit is the catalytic subunit that in vitro cleaves its substrate with and without the protein subunit. LÄS MER

  4. 4. Computational Protein Evolution : Modeling the Selectivity and Promiscuity of Engineered Enzymes

    Författare :Klaudia Szeler; Shina C. Lynn Kamerlin; Mikael Widersten; Maria Ramos; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; enzymes; kinetics; catalysis; density functional theory; empirical valence bond; diesters; evolution; promiscuity; Biokemi; Biochemistry;

    Sammanfattning : Enzymes are biological catalysts that significantly increase the rate of all biochemical reactions that take place within cells and are essential to maintain life. Many questions regarding their function remain unknown. LÄS MER

  5. 5. Quantum Chemical Modeling of Phosphoesterase Mimics and Chemistry in Confined Spaces

    Författare :Henrik Daver; Fahmi Himo; Gregori Ujaque; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; density functional theory; catalysis; phosphoester hydrolysis; transesterification; supramolecular chemistry; inclusion complex; host-guest chemistry; cycloaddition; organisk kemi; Organic Chemistry;

    Sammanfattning : In this thesis, density functional theory is employed in the study of two kinds of systems that can be considered to be biomimetic in their own ways. First, three binuclear metal complexes, synthesized by the group of Prof. Ebbe Nordlander, have been investigated. LÄS MER
