1. Cell signaling by Rho and Miro GTPases : Studies of Rho GTPases in Cytoskeletal Reorganizations and of Miro GTPases in Mitochondrial Dynamics
Sammanfattning : The Ras superfamily of GTPases embraces six major branches of proteins: the Ras, Rab, Ran, Arf, Rho and Miro subfamilies. The majority of GTPases function as binary switches that cycle between active GTP-bound and inactive GDP-bound states. This thesis will focus primarily on the biological functions of the Rho and Miro proteins. LÄS MER
2. Statins, Lipids, and Mutations: Consequences for the Heart and Immune System
Sammanfattning : CAAX proteins are a group of proteins that undergo a three-step protein maturation process that renders the proteins carboxyl-terminus hydrophobic and prone to localize to cellular membranes, where they have their primary function. The first step in this process is called prenylation, which is the covalent attachment of a lipid, either a 15-carbon farnesyl or a 20-carbon geranylgeranyl lipid, to the carboxyl-terminal cysteine residue by the enzymes farnesyltransferase (FTase) and geranylgeranyltransferase type I (GGTase-I), respectively. 3. Defining the Role of GGTase-I in the Development of Rheumatoid Arthritis and Atherosclerosis
Sammanfattning : RHO family proteins control cell movement and intracellular signaling by cycling between active GTP-bound and inactive GDP-bound states. Aberrant signaling of RHO GTPases has been implicated in many diseases including cancer and inflammation. LÄS MER
4. RICH-1, a Multifunctional RhoGAP Domain-containing Protein, Involved in Regulation of the Actin Filament System and Membrane-trafficking
Sammanfattning : The Rho GTPases, which are related to the Ras family of proto-oncogenes, have been found to have important roles in regulating the morphogenic and migratory properties of eukaryotic cells. In addition, these proteins have been shown to regulate aspects of cell signaling, cell growth, cell division and cell survival. LÄS MER
5. Defining the importance of protein geranylgeranylation in innate immunity
Sammanfattning : RHO family proteins and other intracellular proteins are prenylated with a 20-carbon lipid—a product of the cholesterol synthesis pathway—by protein geranylgeranyltransferase type I (GGTase-I). Prenylation is widely believed to target proteins to membranes where they encounter effector molecules that stimulate GTP-binding and activation. LÄS MER
