1. Macrolide Antibiotics in Bacterial Protein Synthesis
Sammanfattning : Macrolides are a large group of clinically relevant antibiotics that inhibit protein synthesis by binding to the large ribosomal subunit in the peptide exit tunnel, close to the peptidyl transferase center (PTC). We have shown that the peptide length of the resulting peptidyl-tRNA drop-off products is proportional to the distance between the PTC and the respective macrolide in the tunnel. LÄS MER
2. Synthesis of Novel Homoserine Analogues - Towards t he Development of Substrate-based Peptide Inhibitors of cAMP-dependent Protein Kinase
Sammanfattning : Protein kinases are enzymes that via phosphorylation of proteins/peptides regulate most aspects of cell functions and physiology. The enzymes transfer a -phosphoryl group from ATP to hydroxyl groups of Ser, Thr and Tyr residues in their substrates. LÄS MER
3. Mechanisms of Adaptation to Deformylase Inhibitors
Sammanfattning : Antibiotic resistance is a growing problem on a global scale. Increasing numbers of bacteria resistant toward one or multiple antibiotics could return us to the high mortality rates for infectious diseases of the pre-antibiotic era. LÄS MER
4. Design, Synthesis and Characterization of Small Molecule Inhibitors and Small Molecule : Peptide Conjugates as Protein Actors
Sammanfattning : This thesis describes different aspects of protein interactions. Initially the function of peptides and their conjugates with small molecule inhibitors on the surface of Human Carbonic Anhydrase isoenzyme II (HCAII) is evaluated. LÄS MER
5. Glycoconjugates : synthesis and investigation of carbohydrate-protein interactions
Sammanfattning : To study the functions of glycoconjugates in biological systems reliable and efficient protocols for glycoconjugate synthesis are needed. To reach this goal we have developed methods for solid-phase synthesis of glycoconjugates that can be monitored with gel-phase 19F spectroscopy using fluorinated linkers, building blocks, and protecting groups.
