1. Islet Xenotransplantation : An Experimental Study of Barriers to Clinical Transplantation
Sammanfattning : In the field of transplantation, the increasing deficit of human donors have lead to an interest in animals as an alternative source of organs and tissues. Different in vitro systems and rodent models of xenotransplantation were used to examine the most significant barriers that have to be overcome, before isolated islets of Langerhans from pigs can be used as a cure for insulin-dependent diabetes mellitus in humans. 2. Studies of Innate and Adaptive Immunity in Islet Transplantation
Sammanfattning : Clinical islet transplantation is today an established alternative treatment for a selected group of type 1 diabetes patients. The predominant technique for transplantation is infusion of islets in the liver via the portal vein. LÄS MER
3. Islet xenograft rejection : studies in the pig-to-rodents and pig-to-primate models
Sammanfattning : The aims of this thesis was: 1) to compare islet allo- and xenograft rejection in rats, using immunosuppression with CsA in a mixed allogeneic-xenogeneic islet transplantation model, 2) to evaluate the efficacy of various immunosuppressive drugs, with special reference to Leflunomide, in preventing islet xenograft rejection in the pig-to-rat model, 3) to perform an immunohistochemical study of porcine islet xenograft rejection in primates including primates treated with CsA+DSG, and 4) to study the metabolism and excretion of porcine C-peptide in mice, with special reference to the use of donor-specific C-peptide for monitoring islet xenograft function. In summary, the results were as follows: 1. LÄS MER
4. Neogenesis and alloxan toxicity in pancreatic islets, with special reference to the transplanted human β-cell
Sammanfattning : The aim of this work was to study whether adult human pancreatic β-cells have the capacity to proliferate in culture and after transplantation to the renal subcapsular space of mice. An immunosuppressive regimen for human islet discordant xenotransplantation to mice was designed. LÄS MER
5. Immunity against porcine islet xenografts in man
Sammanfattning : Due to the lack of organs for clinical transplantation, alternative sources have been investigated. One solution to this shortage could be xenotransplantation, i.e. the transplantation of organs/cells across species barriers. LÄS MER
