1. Mitochondrial medicine. New strategies to evaluate drug toxicity and develop pharmacological protection of the cell’s powerhouse
Sammanfattning : Mitochondria produce the majority of the cell’s energy. Any dysfunction in, or interference with mitochondrial function can have severe consequences. And yet, it was only within the last decades that screening for potential mitochondrial toxicity was included as routine toxicity assay during early drug development. LÄS MER
2. Anti-epileptic and neuroprotective mechanisms by Topiramate and Levetiracetam evaluated in primary cultures
Sammanfattning : The aim of this thesis was to evaluate plausible intracellular mechanisms of action for antiepileptic drugs, Topiramate (TPM) and Levetiracetam (LEV), and to investigate if TPM possesses neuroprotective properties. The thesis is based on results from calcium imaging, immunohistochemical stainings, viability tests, and Western blot using primary cortical or hippocampal cultures, or hippocampal brain slices. 3. Sperm Membrane Channels, Receptors and Kinematics : Using boar spermatozoa for drug toxicity screening
Sammanfattning : Internal fertilization usually implies that a spermatozoon, with intact attributes for zygote formation, passes all hurdles during its transport through the female genitalia and reaches the oocyte. During this journey, millions to billions of other spermatozoa perish. LÄS MER
4. Statin-induced cellular effects mediated by the P2X7 receptor
Sammanfattning : Cholesterol-lowering statins have been shown to inhibit growth of different cancer cells both in vitro and in vivo. Epidemiological studies also indicate a chemopreventive effect of statins. We have investigated the effect of statins on Akt/protein kinase B signaling and their sensitizing effect of cytostatic drugs. LÄS MER
5. High-throughput screening using multicellular tumor spheroids to reveal and exploit tumor-specific vulnerabilities
Sammanfattning : High-throughput drug screening (HTS) in live cells is often a vital part of the preclinical anticancer drug discovery process. So far, two-dimensional (2D) monolayer cell cultures have been the most prevalent model in HTS endeavors. However, 2D cell cultures often fail to recapitulate the complex microenvironments of in vivo tumors. LÄS MER
