1. Recombinant subunit vaccines : protein immunogens
Sammanfattning : Recombinant techniques provide valuable tools for thedevelopment of modern subunit vaccines. In this thesisdifferent systems for production of recombinant subunitvaccines are presented. The strategies investigated includeprotein immunogens, live bacterial vectors and nucleicacids. LÄS MER
2. Structural Studies of Binding Proteins: Investigations of Flexibility, Specificity and Stability
Sammanfattning : Binding proteins are present both in gram-negative and gram-positive bacteria. They are the recognition components of the ABC transport systems that transport different nutrients into the cell, and are in some cases also involved in chemotaxis. LÄS MER
3. The Membrane-Spanning Domain of Complex I Investigated with Fusion Protein Techniques
Sammanfattning : NADH:quinone oxidoreductase, or complex I, is a large, complex and poorly understood bioenergetic enzyme in the respiratory chain of living organisms. The enzyme has a conserved core structure, comprising fourteen protein subunits: Seven subunits protrude from the membrane and contain a flavin prosthetic group and eight iron-sulfur clusters that guide electrons from the oxidation of NADH towards the quinone binding site(s). 4. Structural studies of sugar binding proteins
Sammanfattning : Binding proteins, which are themselves non-enzymatic, play an important role in enzymatic reactions as well as non-enzymatic processes by providing a binding platform for the specific recognition of particular molecules. For example, periplasmic binding proteins play a vital role in nutrient uptake in Gram-negative bacteria. LÄS MER
5. Into the Membrane and Beyond : Improving Membrane Protein Over-Expression in Escherichia coli
Sammanfattning : Membrane proteins fulfil a wide variety of essential functions in the cell and many are (potential) drug targets. Since their natural abundance is usually very low, most membrane proteins have to be over-expressed for functional and structural studies. LÄS MER
