1. Studies of protein structure, dynamics and protein-ligand interactions using NMR spectroscopy
Sammanfattning : In the first part of the thesis, protein-ligand interactions were investigated using the chaperone LcrH, from Yersinia as target protein. The structure of a peptide encompassing the amphipathic domain (residue 278-300) of the protein YopD from Yersinia was determined by NMR in 40% TFE. LÄS MER
2. Conformational Studies of Biologically Active Peptides
Sammanfattning : In this work conformational studies have been performed on peptides from four different systems in an attempt to gain knowledge about their biological function. Circular dichroism (CD) spectroscopy and nuclear magnetic resonance (NMR) spectroscopy in combination with distance geometry and restrained molecular dynamics simulations were the methods used. 3. Synthesis, conformational analysis, and biological evaluation of peptides from E. coli P pilus proteins
Sammanfattning : Many bacteria utilize hairlike protein appendages (pili) to attach themselves to mucosal cell surfaces in the host organism. The pilus is assembled from protein subunits synthesized inside the bacteria and transported to the outer cell wall for incorporation into the growing pilus. LÄS MER
4. Combinatorial Synthesis of Pilicides
Sammanfattning : This thesis describes a combinatorial approach to design and synthesis of three libraries of compounds, referred to as pilicides, designed to target periplasmic chaperones present in uropathogenic Escherichia coli bacteria that are the main cause of urinary tract infections. Periplasmic chaperones are required for assembly of adhesive organelles, i. LÄS MER
5. Antiadhesive agents targeting uropathogenic Escherichia coli : Multivariate studies of protein-protein and protein-carbohydrate interactions
Sammanfattning : This thesis describes studies directed towards development of novel antiadhesive agents, with particular emphasis on compounds that prevent attachment of bacteria to a host-cell. Three different proteins involved in the assembly or function of adhesive pili in uropathogenic Escherichia coli have been targeted either by rational structure based design or statistical molecular methods.
