1. Regulation of IL-6-signaling and inflammatory response – Role of insulin, FOXO1 and PKCδ
Sammanfattning : The aim of this thesis was to investigate if and how insulin antagonizes interleukin-6 (IL-6)-signaling and action using both 3T3-L1 mouse adipocytes (Paper I) and HepG2 human hepatocytes (Paper III). We also investigated the importance of PKCδ in 3T3-L1 adipocytes and mouse embryonic fibroblasts (MEFs) lacking protein kinase C-δ (PKCδ-/-) (Paper II). LÄS MER
2. Roles of protein kinase C in cell death and breast cancer
Sammanfattning : Aberrant protein kinase C (PKC) activity and expression is implicated in different malignancies. To study the role of different PKC isoforms in breast cancer the expression of PCKα, δ and ε was evaluated in breast cancer tumors. In addition the effect of siRNA-mediated knockdown of the isoforms was studied in a global gene expression analysis. LÄS MER
3. Protein Kinase C as an Apoptosis Regulator and a Potential Prognostic Factor in Breast Cancer
Sammanfattning : Several protein kinase C (PKC) isoforms have been suggested as potential targets for breast cancer therapy. To explore the role for PKC isoforms in processes that facilitates malignant progression and the utility of PKC isoforms as biomarkers in breast cancer, we designed a study where we evaluated the expression of PKCα, δ, and ε in primary breast cancers as well as in breast cancer cell lines. 4. Functional and genetic studies in type 2 diabetes and obesity
Sammanfattning : Type 2 diabetes (T2D) and obesity are highly prevalent disorders reflecting a complex interplay of genetics, epigenetics, and environment. They constitute serious health problems and lead to significant morbidity and mortality. LÄS MER
5. The role of PKC in neuronal differentiation and in regulation of the actin cytoskeleton during neurite outgrowth
Sammanfattning : Protein kinase C (PKC) is a family of serine/threonine kinases subgrouped into classical (α, βI, βII, γ), novel (δ, ε, η, θ) and atypical (ζ, ι/λ) isoforms. PKC can be activated by phorbol esters, and prolonged treatment of neuroblastoma cells has been shown to induce neuronal differentiation with neurite outgrowth and increased expression of neuronal genes.
