1. Immunological mechanisms involved in seasonal pollen allergy and allergen-specific immunotherapy
Sammanfattning : Th1/Th2-like T-cell responses were studied by stimulating peripheral blood mononuclear cells (PBMCs) with allergens or antigens followed by restimulation with mitogens to induce high cytokine synthesis. Allergen-stimulation in vitro induced Th2-skewed cytokine synthesis by PBMCs from birch-pollen-allergic patients. LÄS MER
2. The role of Malassezia in the pathogenesis of atopic eczema/dermatitis syndrome
Sammanfattning : Atopic eczema/dermatitis syndrome (AEDS) is a chronically relapsing skin disease, which is often associated with elevated levels of serum lgE. The immunopathology of AEDS is at present unclear, but it is known that lymphocytes infiltrating the AEDS lesions are predominantly of the CD4+ T helper phenotype. LÄS MER
3. Immunomodulation by intravenous immunoglobulin : the effect on cytokine production studied at the single-cell level
Sammanfattning : Preparations of pooled human immunoglobulin for intravenous use (IVIg) have an established therapeutic role in immunodeficiencies, Idiopathic Thrombocytopenic Purpura and Kawasaki's disease and has been attempted in other immunologically mediated conditions including autoimmunity, malignancies, bone marrow transplantation and serious infections such as group A streptococcal toxic shock syndrome. The effects of IVIg on cytokine synthesis in peripheral blood mononuclear cells (PBMC) was studied in vitro by applying a new immunohistochemical method for detection of cytokine production at a single cell level. 4. Evaluation of HIV testing strategies and monitoring of immune responses in HIV-vaccinated individuals in Tanzania
Sammanfattning : This thesis describes studies on the evaluation of human immunodeficiency virus (HIV) enzyme-linked immunosorbent assays (ELISAs) and simple rapid HIV assays for use in HIV testing strategies in resource-limited settings and studies of HIV vaccine-induced immune responses. Peripheral blood mononuclear cell (PBMC) preparation techniques were also studied in preparation for use in the HIV vaccine trials. LÄS MER
5. Innate and acquired immunity to Leishmania in humans : the role of the host versus the parasite
Sammanfattning : Leishmania are intracellular protozoan parasites of macrophages which induce chronic diseases in man. The study of this parasite has been instrumental in elucidating many facets of the immune system. Our study set out to understand the immune regulatory mechanisms involved in resistance and susceptibility. LÄS MER
