1. Determining the effects of regulatory parameters on the structural dynamics of P-type ATPase membrane transporters
Sammanfattning : Proteins are macromolecular machines with roles in all cellular activities and structures. The functional properties of each protein is the result of its combination of 3D-structure and inherent dynamics, and a wealth of structural and dynamic mechanisms have evolved to regulate protein activity. 2. P-type ATPases studied by electron microscopy and two-dimensional crystallization
Sammanfattning : Electron microscopy and two-dimensional crystallography have been used to study the molecular structures of the porcine renal Na+,K+-ATPase and the porcine gastric H+,K+-ATPase. In addition, morphological changes have been observed with electron cryomicroscopy from tubulovesicles in porcine gastric mucosae. LÄS MER
3. Calcium transporting ATPases in plant cells
Sammanfattning : Plant cells carry Ca2+ ATPases to maintain the low resting level of cytosolic Ca2+ (0.1-0.2 µM) found in all eukaryotes. Plant Ca2+ ATPases are mainly located in the plasma membrane, the endoplasmic reticulum and in the vacuolar membrane, and consequently large Ca2+ gradients are created across these membranes. LÄS MER
4. The choreography of protein vibrations : Improved methods of observing and simulating the infrared absorption of proteins
Sammanfattning : The work presented in this thesis has striven toward improving the capability to study proteins using infrared (IR) spectroscopy. This includes development of new and improved experimental and theoretical methods to selectively observe and simulate protein vibrations. LÄS MER
5. The Plasma Membrane H+-ATPase - Identification of a 14-3-3 binding motif
Sammanfattning : The P-type plasma membrane H+-ATPases form a group of proteins only found in plants and fungi. The pumping of protons across the plasma membrane, energized by ATP hydrolysis, creates an electrochemical gradient that is essential for solute transport and internal pH regulation. LÄS MER
