  1. 1. ATP-Binding-Cassette Transporters in Biliary Efflux and Drug-Induced Liver Injury

    Författare :Jenny M. Pedersen; Per Artursson; Christel A. S. Bergström; Stefan Oswald; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; ABC transport protein; Pgp; P-glycoprotein; ABCB1; BCRP; breast cancer resistance protein; ABCG2; MRP2; multidrug resistance-associated protein 2; ABCC2; BSEP; bile salt export pump; ABCB11; sandwich cultured human hepatocytes; SCHH; drug-induced liver injury; DILI; multivariate data analysis; OPLS; Galenisk farmaci; Pharmaceutics;

    Sammanfattning : Membrane transport proteins are known to influence the absorption, distribution, metabolism, excretion and toxicity (ADMET) of drugs. At the onset of this thesis work, only a few structure-activity models, in general describing P-glycoprotein (Pgp/ABCB1) interactions, were developed using small datasets with little structural diversity. 2. 2. Interindividual Variability of Drug Transport Proteins : Focus on Intestinal Pgp (ABCB1) and BCRP (ABCG2)

    Författare :Gunilla Englund; Per Artursson; Håkan Melhus; Andreas Kindmark; Joseph W Polli; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutics; membrane transport proteins; ATP-Binding Cassette Transporters; P-glycoprotein; Breast Cancer Resistance Protein; Interindividual variability; Galenisk farmaci; Pharmaceutics; Galenisk farmaci;

    Sammanfattning : The appearance of adverse drug reactions is a common reason for hospitalization in Western countries. Research on underlying biological mechanisms for interindividual variability in drug response aims to better identify patients with exceptional genetic traits, disease conditions or risk of drug-drug interactions and thereby help to prevent adverse drug reactions. 3. 3. Drug Transport and Metabolism in Rat and Human Intestine

    Författare :Sofia Berggren; Hans Lennernäs; Janet Hoogstraate; Patrick Augustijns; Uppsala universitet; []
    Nyckelord :Biopharmacy; Ussing chamber; microsome; single-pass perfusion; human; rat; intestine; permeability; efflux; P-glycoprotein; metabolism; cytochrome P450; ropivacaine; lidocaine; bupivacaine; Biofarmaci; PPX;

    Sammanfattning : One of the aims of this thesis was to investigate the involvement of efflux proteins, such as the P-glycoprotein (Pgp), in the drug transport in different regions of the rat and the human intestine. The intestinal extrusion of intracellularly formed CYP3A4 metabolites, including whether this extrusion might be mediated by Pgp, was also studied. LÄS MER

  4. 4. Intestinal barriers to oral drug absorption: Cytochrome P450 3A and ABC-transport proteins

    Författare :Helena Engman; Per Artursson; Hans Lenernäs; Heyo K Kroemer; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutics; oral drug delivery; bioavailability; human jejunum; Caco-2; ABC-transporter; P-glycoprotein; CYP3A; Galenisk farmaci; Pharmaceutics; Galenisk farmaci;

    Sammanfattning : The subject of this thesis was to study two intestinal barriers to oral drug bioavailability, drug efflux proteins of the ABC-transporter family, and in particular ABCB1/P-glycoprotein (Pgp), and the drug metabolizing enzyme cytochrome P450 (CYP) 3A4. At the onset of this thesis, similarities between CYP3A4 and Pgp in terms of their tissue distribution and gene regulation, along with overlapping substrate specificities, had generated the hypothesis that CYP3A4 and Pgp may have a complementary function and thus form a coordinated intestinal barrier to drug absorption and gut wall metabolism. 5. 5. Prognostic factors in soft tissue sarcoma. Tissue microarray for immunostaining, the importance of whole-tumor sections and time-dependence

    Författare :Jacob Engellau; Bröstcancer-genetik; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; growth pattern; time-dependence; P-glycoprotein Pgp ; CD44; β-catenin; cyclin A; bcl-2; p53; Ki-67; immunohistochemistry IHC ; whole-tumor sections; tissue microarray TMA ; Soft tissue sarcoma; prognostic factors; Cytology; oncology; cancerology; Cytologi; onkologi; cancer;

    Sammanfattning : In adult soft tissue sarcoma (STS) of the extremities and trunk wall, improved prognostic factors are needed to identify patients at high-risk for metastasis. Various factors are included in the many prognostic systems currently in use and the prognostic value of immunohistochemical (IHC) expression of biological markers is unclear. LÄS MER
