  1. 1. High-throughput screening using multicellular tumor spheroids to reveal and exploit tumor-specific vulnerabilities

    Detta är en avhandling från Uppsala : Acta Universitatis Upsaliensis

    Författare :Wojciech Senkowski; Uppsala universitet.; [2017]
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; spheroids; high-throughput screening; nitazoxanide; OXPHOS; gene expression profiling; statins; drug repositioning; 3D cell culture; quiescent cells; Medical Science; Medicinsk vetenskap;

    Sammanfattning : High-throughput drug screening (HTS) in live cells is often a vital part of the preclinical anticancer drug discovery process. So far, two-dimensional (2D) monolayer cell cultures have been the most prevalent model in HTS endeavors. However, 2D cell cultures often fail to recapitulate the complex microenvironments of in vivo tumors. LÄS MER

  2. 2. The significance of mitochondrial respiratory function in regulating oxygen uptake and performance in humans

    Detta är en avhandling från Stockholm : Gymnastik- och idrottshögskolan, GIH

    Författare :Daniele A. Cardinale; GIH. Gymnastik- och idrottshögskolan; [2018]
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; mitochondria; OXPHOS; p50mito; mitochondrial oxygen affinity; oxygen extraction; sex difference; hyperoxia; training; muscle metabolism; performance; HIIT; Medicin Teknik; Medicine Technology;

    Sammanfattning : The mitochondrion is one of the most fascinating organelles of our cells which has kept and keeps researchers busy in studying its origin, the complex morphology, the numerous functions, the rapid adaptations to a variety of stimuli and its role in health and disease. Exercise challenges cellular homeostasis and skeletal muscle mitochondria greatly adapt to repeated bouts of exercise by increasing mitochondrial respiratory function and content to match energy requirements and to better sustain future perturbations induced by muscle contractions. 3. 3. Modulators of Saccharomyces cerevisiae cytochrome c oxidase Implications for the regulation of mitochondrial respiration

    Detta är en avhandling från Stockholm : Department of Biochemistry and Biophysics, Stockholm University

    Författare :Camilla Rydström Lundin; Stockholms universitet.; [2018]
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; cytochrome c oxidase; cytochrome c; OXPHOS; membrane protein; kinetics; ligand-binding; electron transfer; Rcf1; respiratory supercomplexes; Saccharomyces cerevisiae; biokemi; Biochemistry;

    Sammanfattning : Oxidative phosphorylation in mitochondria is performed by enzyme complexes and electron carriers that reside in the inner membrane. It is now generally accepted that these respiratory enzyme complexes assemble into larger so-called supercomplexes. However, it is presently not known why, under which conditions or how these supercomplexes form. LÄS MER

  4. 4. Mitochondrial function in sepsis - Temporal evolvement of respiratory capacity in human blood cells

    Detta är en avhandling från Department of Clinical Sciences, Lund University

    Författare :Fredrik Sjövall; Lund University.; Lunds universitet.; [2013]
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; human; sepsis; severe sepsis; septic shock; platelets; mitochondria; OXPHOS; high-resolution respirometry; mitochondrial disorders; multiple organ failure; biogenesis; leukocytes; cytokines; respirometry; oxygen consumption; blood cells; temporal;

    Sammanfattning : Sepsis is a devastating disease that is caused by the host’s response to an overwhelming infectious process. As sepsis progresses, organs distant from the site of infection become affected and sepsis-induced multiple organ failure ensues. LÄS MER

  5. 5. Genetic and epigenetic influence on oxidative phosphorylation, islet function and type 2 diabetes in humans

    Detta är en avhandling från Department of Clinical Sciences, Lund University

    Författare :Anders H Olsson; Lund University.; Lunds universitet.; Lund University.; Lunds universitet.; [2012]
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Type 2 Diabetes; Genetics; Epigenetics; Human Pancreatic Islets; Oxidative Phosphorylation; Insulin Secretion;

    Sammanfattning : The prevalence of type 2 diabetes (T2D) is increasing worldwide. T2D is a heterogeneous disease caused by a complex interplay between multiple genetic, epigenetic and non-genetic factors. The disease is characterised by impaired insulin secretion from pancreatic β-cells and insulin resistance in peripheral tissues. LÄS MER
