11. Targeting the hematopoietic stem cell to correct osteopetrosis
Sammanfattning : This thesis focuses on developing stem cell targeted gene therapy for the severe hereditary disorder Infantile Malignant Osteopetrosis (IMO) as well as increasing the understanding of how the genetic defect present in IMO affects the hematopoietic stem cells and hematopoiesis in general. IMO is a rare congenital disorder associated with an increased number of non-functional osteoclasts and the absence of bone resorption results in accumulation of sclerotic bone leading to abnormal bone marrow cavity formation insufficient to support hematopoiesis. 12. Small-scale dosimetry methods for normal tissues after radioembolization and peptide receptor radiotherapies; a focus on the liver parenchyma and the bone marrow
Sammanfattning : Radionuclide therapies are increasingly used in cancer treatments as they have been proven to be both effective and safe. As biodistributions and uptake vary widely between patients, a standardized approach is bound to leave a fraction of patients overtreated or undertreated. LÄS MER
13. Cellular and Molecular Pathways Governing Hematopoietic Stem Cell Fate
Sammanfattning : Hematopoietic Stem Cells (HSCs) have been defined to have abilities of both self-renewal and multi-lineage differen-tiation. The balance between these abilities is rigorously controlled under steady-state status (homeostasis) and when the normal processes of self-renewal and differentiation become deregulated, a disorder of blood system (such as leukemias) can occur. 14. CD47–SIRPα : an interaction of importance for bone cell differentiation
Sammanfattning : Bone tissue is continuously remodeled by bone-forming osteoblasts and bone-resorbing osteoclasts, in processes tightly regulated by hormones, cytokines and growth factors. CD47, a ubiquitously expressed protein, and one of its ligands, signal-regulatory protein alpha (SIRPα), are two cell-surface proteins belonging to the immunoglobulin (Ig)-superfamily. LÄS MER
15. Targeted radionuclide therapy for patients with neuroendocrine tumours with focus on normal tissue response in 177-Lu-DOTATATE treatment
Sammanfattning : Targeted radionuclide therapy with 177Lu-DOTATATE for patients with neuroendocrine tumours utilises the frequent overexpressing of somatostatin receptors on the tumour cells. This treatment modality has demonstrated valuable patient benefits and is well tolerated. However, renal and bone marrow toxicity can become dose limiting and persisting. LÄS MER
