2. Temporal events in neuronal differentiation and cell death : expression and processing of the Alzheimer's amyloid precursor protein family and a protein at the nuclear pore
Sammanfattning : The present study had two major objectives: 1) to elucidate the involvement of Alzheimer’s amyloid precursor protein (APP) family in neuronal differentiation, and the effect of the Alzheimer’s disease (AD)-linked mutation APPV642I on signal transduction; 2) to investigate the fate of the nuclear pore complex protein POM121 during apoptosis and to examine the possibility of using green fluorescent protein (GFP)-labelled POM121 as a non-invasive sensor of apoptosis in living (non-fixed) cells.APP is the parent protein of the b-amyloid peptide, which is the major peptide constituent in the “senile plaques” of AD. Sammanfattning : In the central nervous system (CNS) dopaminergic and dopaminoceptive neurons have the ability to respond to variations in dopamine levels by for example adjusting their dopamine receptor levels. One of the most well known phenomena in this respect is that long-term blockade of D2 receptors with antipsychotic drugs leads to an increase in striatal D2 receptor density of experimental animals as well as of schizophrenic patients as measured in both post-mortem studies and in vivo positron emission tomography studies of brain. Sammanfattning : A crucial aspect of drug delivery is efficient transport to the site of action. Thus, there is a need to design and evaluate new delivery vehicles. In this thesis two delivery vehicles, cell-penetrating peptides and bacterial ghosts, were evaluated. LÄS MER
Sammanfattning : The proinflammatory cytokines interleukin (IL) -1α, IL-1β and IL-6 are key mediators in the host's response to injury and infection. One of the systemic responses elicited by these proinflammatory cytokines, when injected peripherally or centrally, is fever. Thus, these proteins may act as endogenous pyrogens. LÄS MER
