Sammanfattning : Non-competitive NMDA receptor antagonists, such as phencyclidine (PCP) and dizocilpine (MK-801) can even acutely induce a drug induced psychotic state which closely resembles schizophrenia. The midbrain dopamine (DA) neurons, which originate in the ventral tegmental area (VTA), are involved in motivational, attentional and cognitive processes, and have also been profoundly implicated in schizophrenia. 3. Effects of ethanol on NMDA receptor-mediated functions in primary cultures of cerebellar granule cells
Sammanfattning : Primary cultures of cerebellar granule cells were used to investigate the effects of acute and chronic exposure of ethanol on NMDA receptor functions and to examine the possible sites of interaction between ethanol and the NMDA receptor. Activation of NMDA receptors induced elevation of free cytoplasmic Ca2+, Ca2+ influx, activation of protein kinase C and neurotoxicity. LÄS MER

Sammanfattning : Pain perception and the response to opioid analgesics are determined by a number of factors, among which genetic variability and the activation of the NMDA-receptor for glutamate play an important role. The effects of two clinically available NMDA-receptor antagonists, ketamine (KET) and dextromethorphan (DEX), were evaluated on an animal model of acute nociception, the hot plate test, and on human experimental acute ischemic pain. 5. Regulation of NMDA receptor properties by amino acids and cations : a biochemical and electrophysiological study
Sammanfattning : NMDA receptors are glutamate-regulated ion channels that are permeable to Ca2+, Na+ and K+ and are sensitive to voltage-dependent Mg2+-block. To investigate the modulatory effects of amino acids and cations on NMDA receptors and the possible regional differences in the pharmacology of NMDA receptors, I used mainly two techniques in this thesis: electrophysiological analysis of expressed NMDA receptors in Xenopusoocytes and biochemical receptor binding using [3H]MK-801 in membrane preparations from the rat cerebral cortex and spinal cord.
