An understanding of what happens at the cellular and molecular level in prostate cancer cells, while reducing their access to androgens and then exposing them to ionizing radiation (IR), would give us an opportunity to optimize the treatment and may also inspire novel therapeutic approaches. We found that the NHEJ protein Nej1 interacted physically with the HR protein Srs2, which was dependent on phosphorylation of Nej1 by Dun1. Srs2 recruitment to a DSB partly relied on Nej1 and Dun1. This includes transportation of the Survival of motor neuron (SMN) complex, small Cajal body-specific (sca) RNAs and the telomerase enzyme to Cajal bodies, as well as targeting of the E3 ligase RNF8 to DNA double-strand breaks (DSBs).
