  1. 1. Engineering Antibody Specificity Using Combinatorial Antibody Libraries

    Författare :Jenny Boström; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; antibody Antigen Interactions; Dual Specific Antibodies; Herceptin; Human Epidermal Growth Factor Receptor 2; Thermodynamic Analysis; therapeutic Antibody Engineering; Synthetic Phagedisplayed Antibody Libraries; Vascular Endothelial Growth Factor; Bacterial Display; trastuzumab;

    Sammanfattning : The research presented in this thesis investigates antibody specificity, antibody-antigen and protein-protein interactions using combinatorial antibody libraries, phage-display and bacterial display technology. One goal was to develop a method to generate antibodies that bind two protein antigens with high affinity using a single-antigen binding site, and to investigate the molecular and thermodynamic requirements for antibody multispecificity. 2. 2. Development and evaluation of analytical techniques for antibodies and antibody-drug conjugates : From verification of conjugation to stability testing

    Författare :Malin Källsten; Jonas Bergquist; Fredrik Lehmann; Sara Lind; Lucia Kovac; Alain Beck; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Antibody-drug conjugates; drug-to-antibody ratio; monoclonal antibody; mass spectrometry; hydrophobic interaction chromatography; sample preparation; aggregation; Kemi med inriktning mot analytisk kemi; Chemistry with specialization in Analytical Chemistry;

    Sammanfattning : Antibody-drug conjugates (ADCs) are becoming increasingly important in oncology. ADCs are inherently heterogeneous analytes. For a successful drug candidate to go through to the clinic, extensive characterization and evaluation of the constructs are necessary. LÄS MER

  3. 3. Targeted Therapy of Colorectal Cancer : Preclinical Evaluation of a Radiolabelled Antibody

    Författare :Ylva Almqvist; Anders Sundin; Vladimir Tolmachev; Hans Lundqvist; Matthew K Robinson; Uppsala universitet; []
    Nyckelord :Molecular medicine; tumour targeting; antibody; A33; radionuclide; colorectal cancer; therapy; Molekylärmedicin;

    Sammanfattning : Targeted radiotherapy (TRT) of cancer is a promising approach that enables selective treatment of tumour cells, while sparing normal tissue. The humanized monoclonal antibody A33 (huA33) is a potential targeting agent for TRT of colorectal cancer, since its antigen is expressed in more than 95 % of all colorectal carcinomas. LÄS MER

  4. 4. Alpha-Synuclein Oligomers : Cellular Mechanisms and Aspects of Antibody Treatment

    Författare :Gabriel Gustafsson; Martin Ingelsson; Joakim Bergström; Anna Erlandsson; Tiago F Outeiro; Marina Romero-Ramos; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Parkinson s Disease; Alpha-synuclein; Aggregation; Oligomers; Monoclonal Antibody; Glia; Astrocyte; Immunofluorescence; Medicinsk vetenskap; Medical Science;

    Sammanfattning : In Parkinson’s disease (PD) and dementia with Lewy bodies (DLB), aggregated α-synuclein deposit inside cells within the brain. Smaller soluble α-synuclein aggregates, oligomers, are present both intra- and extracellularly. LÄS MER

  5. 5. Immunomodulatory Therapy of Solid Tumors : With a Focus on Monoclonal Antibodies

    Författare :Linda Sandin; Thomas Tötterman; Mangsbo Sara; Fredrik Eriksson; Loskog Angelica; Ola Winqvist; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; in situ checkpoint blockade; antibody-mediated tumor immunotherapy; CTLA-4; CD40; monoclonal antibodies; experimental animal model; Fc gamma receptor; Medical Science; Medicinsk vetenskap;

    Sammanfattning : Cancer, historically considered a genetic disease, is currently acknowledged to affect the whole body. Our immune system is one key player that can elicit a response against malignant cells but can also promote tumorigenesis. Tumors avoid immune recognition by creating a suppressive microenvironment and inducing tolerance. LÄS MER
