  1. 11. A Chemical Approach Toward the Therapeutic Optimization of Cytotoxic Bioconjugates

    Författare :Rafael Hartmann; Luke R. Odell; Fredrik Lehmann; Mats Larhed; Dario Neri; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Chemotherapy; antibody-drug conjugates; organic synthesis; bioorganic chemistry; synthetic method development; analytical method development; Chemistry with specialization in Bioorganic Chemistry; Kemi med inriktning mot bioorganisk kemi; Medicinal Chemistry; Läkemedelskemi; Chemistry with specialization in Organic Chemistry; Kemi med inriktning mot organisk kemi;

    Sammanfattning : The delivery of cytotoxicity to specific cells was recognized to be the key to the effective, chemical treatment of infectious and neoplastic diseases well over a century ago. In the context of oncology, the primary therapeutic challenge is the distinction between healthy and pathological cells despite their pronounced pheno- and genotypic similarity. LÄS MER

  2. 12. Ligands of the Angiotensin II Type 2 Receptor : Exploring structure and function of the AT2R ligand C38

    Författare :Rebecka Isaksson; Mats Larhed; Annette Bayer; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Angiotensin II type 2 receptor; AT2R antagonists; sulfonyl carbamates; bicyclic amides; metabolic stability; functional activity assay; pharmacological profile; medicinal chemistry; structure-activity relationship; Medicinal Chemistry; Läkemedelskemi;

    Sammanfattning : The renin-angiotensin-aldosterone-system (RAAS) control blood-pressure regulation, exerted by the main effector peptide angiotensin II (AngII) binding the angiotensin II type 1 receptor (AT1R). While hypertension is the most known disease caused by over-activity in RAAS, several proteins in the system exhibit protective functions. LÄS MER

  3. 13. Design and synthesis of inhibitors that target the serine protease thrombin, the malarial aspartyl proteases plasmepsin I and II, and the hepatitis C virus NS3 serine protease

    Författare :Per-Ola Johansson; Mats Larhed; Linköpings universitet; []
    Nyckelord :NATURAL SCIENCES; NATURVETENSKAP;

    Sammanfattning : This thesis addresses the design, synthesis, and structure-activity relationships of protease inhibitors that target the serine protease thrombin, the malarial aspartic proteases plasmepsin I and II, and the hepatitis C virus (HCV) NS3 serine protease. Furthermore, the backgrounds of each of the three types of diseases in question are discussed in detail, and consideration is given as to why it is assumed that inhibition of the mentioned proteases will help prevent or cure cardiovascular diseases, malaria, and hepatitis C. Sammanfattning : Tuberculosis (TB) is a major infectious disease, killing about 2 million people annually throughout the world. Today's TB treatment is a lengthy procedure involving a combination of antibiotics. No new TB drug has been introduced onto the market in the past 40 years, and the emergence of multi- and extensively drug-resistant TB calls for new drugs. LÄS MER

  5. 15. Antiadhesive agents targeting uropathogenic Escherichia coli : Multivariate studies of protein-protein and protein-carbohydrate interactions

    Författare :Andreas Larsson; Jan Kihlberg; Fredrik Almqvist; Mats Larhed; Umeå universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Organic chemistry; Antibacterial; pili; antiadhesive agents; structure based design; statistical molecular design; 2-pyridone; amino acid derivative; galabiose; enaminone; SPR; ELISA; QSAR; Organisk kemi; Organic chemistry; Organisk kemi; organisk kemi; Organic Chemistry;

    Sammanfattning : This thesis describes studies directed towards development of novel antiadhesive agents, with particular emphasis on compounds that prevent attachment of bacteria to a host-cell. Three different proteins involved in the assembly or function of adhesive pili in uropathogenic Escherichia coli have been targeted either by rational structure based design or statistical molecular methods.
