  1. 1. Application of Mixed-Effect Modeling to Improve Mechanistic Understanding and Predictability of Oral Absorption

    Författare :Martin Bergstrand; Mats O Karlsson; Hans Ericsson; Bart Ploeger; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Absorption; magnetic marker monitoring; drug release; IVIVC; pharmacometrics; NONMEM; model diagnostics; pcVPC; pvcVPC; visual predictive check; VPC; BQL; paracetamol; sulfapyridine.; Other pharmacy; Övrig farmaci; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi;

    Sammanfattning : Several sophisticated techniques to study in vivo GI transit and regional absorption of pharmaceuticals are available and increasingly used. Examples of such methods are Magnetic Marker Monitoring (MMM) and local drug administration with remotely operated capsules. LÄS MER

  2. 2. Mechanism-based modeling of biological processes involved in oral absorption

    Författare :Benjamin Guiastrennec; Mats O. Karlsson; Martin Bergstrand; Jörg Lippert; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; absorption; bile acids; enterohepatic circulation; food effect; gallbladder; in vitro in vivo correlation; NONMEM; pediatrics; pharmacometrics; tuberculosis; Pharmaceutical Science; Farmaceutisk vetenskap;

    Sammanfattning : For orally administered drugs, the rate and extent of absorption are governed by the physiology of the gastrointestinal tract, the characteristics of the dosage form and the physico-chemical properties of the drug. This thesis primarily aimed to improve the mechanistic understanding and the predictability of processes involved in the absorption of orally administered drugs using a population modeling approach. 3. 3. Modelling and Simulation to Improve Antimalarial Therapy

    Författare :Jesmin Permala Lohy Das; Mats Karlsson; Martin Bergstrand; Joel Tarning; Mads Krielgaard; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacometrics; pharmacokinetics; pharmacodynamics; malaria; artemisinin; weekly dosing; resistant; pregnant populations; intermittent preventive therapy; parasite clearance; day 3 positivity; nomogram; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi;

    Sammanfattning : The introduction of artemisinin-based combination therapy (ACT) substantially reduced malaria-related mortality and morbidity during the past decade. Despite the widespread use of ACT, there is still a considerable knowledge gap with regards to safety, efficacy and pharmacokinetic properties of these drugs, particularly in vulnerable populations like children and pregnant women. Sammanfattning : My thesis addresses the antagonism between the mystery of life and the destructive pursuit of power which forms a central theme in the works of Per Olof Sundman (1922–1992). The purpose is to show that Sundman uses the theme of the encounter/non-encounter and the destructive leader to examine the conditions for allowing a deeper experience of reality and the destructive exercise of power.
