1. Role of MYCN in retinoblastoma : From carcinogenesis to tumor progression
Sammanfattning : Retinoblastoma, a pediatric malignancy of the retina, is primarily driven by the bi-allelic inactivation of the RB1gene. However, a subset of cases are characterized by proficient RB1 functions but with MYCN copy number mutations, suggesting an alternative oncogenic mechanism in the absence of RB1 mutations. LÄS MER
2. Chromosome dynamics and genomic instablity in neuroblastoma. Three genomic pillars: MYCN amplification, numerical and structural changes
Sammanfattning : In this thesis, the main focus has been on the childhood cancer neuroblastoma, one of the most common and lethal childhood tumours. Neuroblastoma has througout the years continued to be a clinical and biological enigma. LÄS MER
3. Polyamine analogues inhibit neuroblastoma cell growth
Sammanfattning : Abstract Neuroblastoma is a highly malignant neoplasm found in children. Although children with high-risk neuroblastoma respond to chemotherapy, relapses are common and an estimated long-time survival prognosis is approximately 15%. Thus, there is a tremendous need for new chemotherapeutic drugs for the treatment for neuroblastoma. LÄS MER
4. Hypoxia and differentiation in human neuroblastoma cells
Sammanfattning : The childhood tumour neuroblastoma is derived from immature cells of the sympathetic nervous system, which have become arrested at different maturation stages. Neuroblastoma is a malignancy with a high degree of heterogeneity, and there is a correlation between a poor differentiation status and a more aggressive phenotype. LÄS MER
5. Models of Retinal Development and Disease
Sammanfattning : For a model of a human disease to be valid and useful, it is important that key genotypic and phenotypic traits are shared between model system and human. The work in this thesis has been focused on generating new and characterizing spontaneous models of three genetic disorders affecting the retina: retinoblastoma, a childhood cancer with its origin in the fetal retina, Stargardt disease, a juvenile form of macular degeneration, and Bardet-Biedl syndrome, a pleiotropic ciliopathy featuring retinal degeneration.
