1. Neuronal and glial differentiation of expanded neural stem and progenitor cells; in vitro and after transplantation
Sammanfattning : In this thesis we have used cells dissected from the lateral ganglionic eminence (LGE), the medial ganglionic eminence (MGE), and the cortical primordium of the embryonic mouse forebrain. The tissue was dissected from either i) wild-type mice, ii) green fluorescent protein (GFP)-, or iii) Gtv-a-expressing transgenic mice, and subsequently grown and expanded in vitro using two different protocols. 2. EMBRYONIC GLIA AND FOREBRAIN NEUROGENESIS; in vitro generation and differentiation of telencephalic neurons
Sammanfattning : The current thesis is aimed to increase our understanding of the generation and differentiation of telencephalic neurons, with emphasis on the role of glial cells in these processes. Using in vitro methods we have studied the role of glial cells in neurogenesis, inheritance of genetic regional specification, and influence on differentiation of neurons. LÄS MER
3. Control of neuronal survival, migration and outgrowth by GDNF and its receptors
Sammanfattning : Glial cell line-derived neurotrophic factor (GDNF) is the prototypical member of a family of growth factors that are indispensable in nervous system development and maintenance. GDNF signals by binding to a multi-component receptor complex comprised of the ligand-binding subunit GFRα1 and the signaling subunit RET or NCAM. LÄS MER
4. GDNF and its receptors : a structure-function analysis of the NCAM-GDNF-GFRalpha1 complex and an examination of potential receptor candidates
Sammanfattning : Neurotrophic factors are small, secreted proteins that are important players in the development and maintenance of the nervous system. Their functions span from neuronal survival and migration to neurite outgrowth and axon guidance, and in some cases even programmed cell death. LÄS MER
5. Specification of Dorsal and Intermediate Telencephalic Character
Sammanfattning : The telencephalon is the most highly evolved region of the vertebrate central nervous system (CNS). The major structures of the telencephalon - the cortex and basal ganglia – derive from the dorsally positioned pallium and the ventrally positioned subpallium, respectively. LÄS MER
