    Sammanfattning : The human glucose transporter GLUT1 is abundant in red blood cells, the blood-brain barrier and epithelial cells, where it mediates the transport of the energy metabolite, glucose. In the present work some properties of GLUT1, including affinity binding of both substrates and inhibitors, transport rates as well as permeabilities of aromatic amino acids and drug-membrane interactions were analyzed by chromatographic methods. 2. 2. Chromatographic Studies of Solute Interactions with Immobilized Red Blood Cells and Biomembranes

    Författare :Ingo Gottschalk; Irving W. Wainer; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Affinity; Binding; Biomembrane; Biotin; Chromatography; Cytochalasin B; Dissociation constant; Drug absorption; Equilibrium; Glucose; GLUT1; Immobilization; Immobilized biomembrane affinity chromatography; Immobilized liposome chromatography; Interaction; Liposome; Membrane protein; Membrane vesicle; Partitioning; Phospholipid bilayer; Proteoliposome; Quantitative; Red blood cell; Solute; Specific; Streptavidin; Wheat germ agglutinin; Biokemi; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Sammanfattning : Specific and non-specific interactions of solutes with immobilized biomembranes were studied using chromatographic methods. Liposomes, proteoliposomes and red blood cell (RBC) membrane vesicles were immobilized by a freeze-thawing procedure, whereas whole RBCs were adsorbed in the gel beds using electrostatic interaction, binding to wheat germ agglutinin (WGA) or the streptavidin-biotin interaction. 3. 3. Applications of hydrodynamic forces for membrane chromatography

    Författare :Björn Johansson Fast; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Membrane chromatography; FRAP; Protein accumulation; Supported lipid bilayer; Microfluidics; SEEC; Diffusion; Hydrodynamic forces; Label-free;

    Sammanfattning : Membrane proteins are proteins that reside in the cell membrane. Due to their position in the cell membrane, the barrier between the cell interior and exterior, they are vital to cell signaling. Moreover, the presence of membrane proteins in the cell's signaling pathways make membrane proteins important drug targets. LÄS MER

  4. 4. On-line Sample Pretreatment Based on Membrane Enrichment, Mainly Using Supported Liquid Membranes, for Chromatographic Analysis

    Författare :Yin Shen; Centrum för analys och syntes; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biological Sample; Environmental Sample; Capillary Gas Chromatography; Ion Chromatography; Automation; Sample Pretreatment; Enrichment; Membrane; Extraction; Analytical chemistry; Analytisk kemi;

    Sammanfattning : The main objective of this work was to construct practical analytical systems for the analysis of environmental and biological samples, using membrane-based techniques for sample pretreatment and chromatography for final analysis. Two types of membrane processes have been applied, with emphasis on automation. LÄS MER

  5. 5. Perfusion Chromatography of membrane proteins

    Författare :Margrit Roobol-Bóza; Himadri Pakrasi; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; biokemi; Biochemistry;

    Sammanfattning : According to genomics up to 40% of all eukaryotic proteins are membrane bound. Their isolation from the biomembrane is essential for structural and functional studies. The major task in membrane protein isolation is obtaining a pure and homogeneous population, while maintaining biological activity, native conformation, and keeping cofactors bound. LÄS MER
