  1. 1. Characterization of parameters influencing intracellular bioavailability and prediction of intracellular drug exposure

    Författare :Andrea Treyer; Per Artursson; Gert Fricker; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; intracellular drug bioavailability; unbound drug concentration; drug disposition; ADME; drug transport; drug metabolism membrane partitioning; phospholipid; drug-drug interaction; antisense oligonucleotide; cell-penetrating peptide; Farmaceutisk vetenskap; Pharmaceutical Science;

    Sammanfattning : This thesis work investigates factors influencing intracellular drug disposition. An experimental method for measurement of intracellular bioavailability (Fic), was used throughout. Fic is defined as the ratio between the unbound drug concentration inside the cell and the compound concentration in the cell exterior. LÄS MER

  2. 2. Phosphatidylinositol 4-kinases in plasma membranes and Affinity partitioning of liposomes: a model system for membrane purification

    Författare :Lars Ekblad; Biokemi och Strukturbiologi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; metabolism; Biokemi;

    Sammanfattning : Polyphosphoinositides, the different phosphorylated variants of the phospholipid phosphatidylinositol (PtdIns), are used in a wide array of intracellular mechanisms in eukaryotic cells. The phosphorylation of PtdIns at carbon D4, catalyzed by phosphatidylinositol 4-kinase, is an important step in the synthesis of several of these molecules. LÄS MER

  3. 3. Enterostatin - target proteins and intracellular mechanisms. Function in food intake and energy metabolism

    Författare :Karin Berger; Molekylär endokrinologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; ATP; energy metabolism; insulin; uncoupling protein; beta-casomorphin; opiate; SK-N-MC; INS-1; Medicin människa och djur ; Medicine human and vertebrates ; fat intake; F1F0-ATP synthase; appetite; enterostatin; satiety; obesity;

    Sammanfattning : Hunger and satiety are the results of complex neural events that involve several neurotransmitters and peptides. Enterostatin is an appetite-regulating peptide released in the intestine in response to fat ingestion. Enterostatin specifically decreases fat intake, but has also metabolic effects like inhibition of insulin secretion. LÄS MER

    Sammanfattning : Pathogenic Yersinia cause a manifold of diseases in humans ranging from mild gastroenteritis (Y. pseudotuberculosis and Y. enterocolitica) to pneumonic and bubonic plague (Y. LÄS MER

  5. 5. Proislet Amyloid Polypeptide (proIAPP) : Impaired Processing is an Important Factor in Early Amyloidogenesis in Type 2 Diabetes

    Författare :Johan F. Paulsson; Gunilla Westermark; Christer Betsholtz; Linköpings universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biosynthesis amyloid; Genetics amyloid; Metabolism amyloid; Islets of Langerhans; Proprotein convertases; Posttranslation protein processing; Medical cell biology; Medicinsk cellbiologi;

    Sammanfattning : Amyloid is defined as extracellular protein aggregates with a characteristic fibrillar ultra-structure, Congo red affinity and a unique x-ray diffraction pattern. At present, 25 different human amyloid fibril proteins have been identified, and amyloid aggregation is associated with pathological manifestations such as Alzheimer’s disease, spongiform encephalopathy and type 2 diabetes.
