  1. 1. Development of Gene Therapy for Hematopoietic Stem Cells using Lentiviral Vectors

    Författare :Niels-Bjarne Woods; Andreas Ooka; Stefan Karlsson; Avdelningen för molekylärmedicin och genterapi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; embryonic stem cell; Hematopoietic stem cell; lentiviral vector; transduction efficiency; transgene expression; multiple vector copy integration; insertional mutagenesis; NOD SCID mouse; cytogenetics; Genetics; extracellulära vätskor; extracellular fluids; cytogenetik; Genetik; Haematology; Hematologi; Lentivirus : genetics; Human; Genetic Vectors; Hematopoietic Stem Cells : metabolism; Animal; Gene Therapy; Leukemia : therapy; Leukemia : metabolism;

    Sammanfattning : Hematopoietic stem cells are an ideal target for genetic manipulation for the purpose of curing hematological disorders as they have the ability to develop into all blood lineages and to self-renew. In this study we demonstrate that lentiviral vectors, based on HIV-1, can efficiently transfer genes into human hematopoietic progenitor and stem cells as assessed following stem cell transplantation in immune compromised mice. 2. 2. Molecular analysis of Streptococcus pyogenes and its interactions with the human host

    Författare :Andreas Berge; Institutionen för experimentell medicinsk vetenskap; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; complement activation; fibrinogen; IgG; M protein; C5a peptidase; streptococcal cysteine proteinase; streptokinase; t-PA; Streptococcus pyogenes; plasminogen; C1q; insertional sequence.; Microbiology; bacteriology; virology; mycology; Mikrobiologi; bakteriologi; virologi; mykologi;

    Sammanfattning : Streptococcus pyogenes causes infections only in humans, from trivial tonsillitis to life-threatening conditions. One of its most important virulence factors is the M-like protein family conferring resistance to phagocytosis to the bacterium. The M-like proteins are surface exposed rod-like molecules attached to the cell wall. Each strain of S. LÄS MER

  3. 3. Screening for Candidate Brain Tumor Genes : Identifying Genes that Cooperate with Platelet-Derived Growth Factor in Glioma Development and Progression

    Författare :Fredrik Johansson; Bengt Westermark; Leif Carlsson; Uppsala universitet; []
    Nyckelord :Pathology; PDGF; insertional mutagenesis; retroviral tagging; common insertion site; glioma; expression; cGMP-dependent protein kinase II; Patologi;

    Sammanfattning : Malignant primary brain tumors, gliomas, often overexpress both platelet-derived growth factor (PDGF) ligands and receptors providing an autocrine and/or paracrine boost to tumor growth. Glioblastoma multiforme (GBM) is the most frequent glioma. Its aggressive and infiltrative growth renders it extremely difficult to treat. LÄS MER

  4. 4. The Entheses of the Medial Meniscns and Ligaments of the Knee Joint : Descriptive and Experimental Studies

    Författare :Jizong Gao; Michael Benjamin; Linköpings universitet; []
    Nyckelord :knee joint; meniscus; ligaments; insertion; enthesis; quantitative morphology; fibrocartilage; cement line; collagens; proteoglycans; nerve fibres; tensile strength; failure mode; transection; refixation; repair; healing; osteoarthrosis; histology; immunohistochemistry; biomechanics; rabbit model; MEDICINE; MEDICIN;

    Sammanfattning : Meniscal injuries are common in sports. Surgical removal of the meniscus to relieve symptoms is associated with a high risk for late knee joint osteoarthrosis and pain. Meniscal repair or replacement has therefore been advocated. LÄS MER

  5. 5. Marginally hydrophobic transmembrane α-helices shaping membrane protein folding

    Författare :Tuuli Minttu Virkki de Marothy; Arne Elofsson; William Dowhan; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; membrane protein folding; hydrophobicity; translocon; transmembrane helix; marginally hydrophobic transmembrane helices; orientational preference; positive inside rule; aquaporin 1; Biochemistry; biokemi;

    Sammanfattning : Most membrane proteins are inserted into the membrane co-translationally utilizing the translocon, which allows a sufficiently long and hydrophobic stretch of amino acids to partition into the membrane. However, X-ray structures of membrane proteins have revealed that some transmembrane helices (TMHs) are surprisingly hydrophilic. LÄS MER
