  1. 11. Cytokine regulation in rodents with experimental arthritis

    Författare :Åsa Boström Müssener; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :collagen-induced arthritis; oil-induced arthritis; IL-2; IL-4; TNFa; IFNy; TGFß;

    Sammanfattning : The general aim of this project has been to try to elucidate the mechanisms behind induction of arthritis by using in situ hybridization and immunohistochemical methods to analyse the dynamics of the cytokine network in vivo, in the peripheral Iymphoid system and in the joints of rodents with collagen-induced arthritis (CIA)and oil-induced arthritis (OIA). We first examined the kinetics of cytokine gene expression in vivo, in the draining Iymph nodes of DA rats following immunization with rat type II collagen (RCII) emulsified in Freund's incomplete adjuvant oil (FIA) or with only FIA, which are regimens that induce CIA and OIA, respectively. 2. 12. Cholesterol turnover in acute myelogenous leukemia with special emphasis on regulation of low density lipoprotein receptor expression in leukemic cells

    Författare :Loukas Tatidis; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Cellular cholesterol requirements are met by receptor mediated uptake of cholesterol in low density lipoprotein (LDL) particles and/or by endogenous synthesis, the rate limiting enzyme in the synthesis pathway being 3-hydroxy-3-methylglutary coenzyme A (HMG-CoA) reductase. Expressions of both the LDL receptor and HMG-CoA reductase genes are normally under end product repression by cellular cholesterol. LÄS MER

  3. 13. Novel immunotherapies for metastatic melanoma - from mouse models towards clinical trials

    Författare :Elin Forsberg; Göteborgs universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Metastatic melanoma; uveal melanoma; patient-derived xenograft; immune-humanized mouse model; immunotherapy; adoptive T cell therapy; tumor-infiltrating lymphocytes; chimeric antigen receptor T CAR-T cells;

    Sammanfattning : Immunotherapies including checkpoint blockade and adoptive T cell transfer (ACT) show great promise for the treatment of melanoma, with long-term effects in some patients. However, around half of the patients with metastatic malignant melanoma will not be cured with available therapies today, and these patients require other treatment strategies. LÄS MER

  4. 14. Activation of murine cytotoxic cells with interelukin-2 and the bacterial superantigen staphylococcal entertoxin A

    Författare :Hans Belfrage; Lund Kirurgi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Immunologi; transplantation; serology; Immunology; cytokine; tolerance; LAK; NK; serologi; CTL; Staphylococcal enterotoxin A; SEA; Interleukin-2; murine; deletion; anergy;

    Sammanfattning : Natural killer (NK) cells and T lymphocytes are the major effector cells that can recognize and kill tumor cells. NK cells have a constitutive cytotoxic activity and kill a wide spectrum of tumor cells while T cells recognize specific tumor antigens and need to be activated through their TCR to differentiate into cytotoxic T lymphocyte (CTL) killers or T helper cells. 5. 15. Effects of mercury and fluoride on human immune cells : elucidation of mechanisms

    Författare :Annika Loftenius; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :human; mercury; fluoride; HgCl2; lymphocyte; superantigen; T-cell receptor; TCRBV dental amalgam;

    Sammanfattning : Mercury, from dental amalgam, and fluoride, administered to prevent dental caries, have been argued to cause adverse health effects. The aims of this thesis have been to examine in what manner the fluoride and mercuric ions interact with human immune cells and, in the case of mercury, if it would be possible to monitor a tentative in vivo effect by in vitro assays.
