1. Antibody Evolution and Repertoire Development
Sammanfattning : Antibodies are key players of the immune system in higher vertebrates, which provide a defense against potentially lethal threats from the environment. Besides their importance in the immune defense, antibodies have a great potential as reagents in biological chemistry and diagnostics, and as therapeutic agents against both infectious diseases and cancer. 2. Generation and evolution of human antibody repertoires
Sammanfattning : This thesis, which is based on five original papers, covers different aspects of antibody specificity and diversity, with the overall goal to understand the makeup of antibody repertoires and their functionality as they occur in vivo or as they are developed in the laboratory. Besides their importance in the adaptive immune response, antibodies have emerged as an invaluable tool in applications ranging from basic research to disease diagnostics and therapy. LÄS MER
3. Engineering Antibody Specificity Using Combinatorial Antibody Libraries
Sammanfattning : The research presented in this thesis investigates antibody specificity, antibody-antigen and protein-protein interactions using combinatorial antibody libraries, phage-display and bacterial display technology. One goal was to develop a method to generate antibodies that bind two protein antigens with high affinity using a single-antigen binding site, and to investigate the molecular and thermodynamic requirements for antibody multispecificity. 4. Biomarkers for Better Understanding of the Pathophysiology and Treatment of Chronic Pain : Investigations of Human Biofluids
Sammanfattning : Chronic pain affects 20 % of the global population, causes suffering, is difficult to treat, and constitutes a large economic burden for society. So far, the characterization of molecular mechanisms of chronic pain-like behaviors in animal models has not translated into effective treatments. LÄS MER
5. Human cytokeratins : their use as targets in cancer management
Sammanfattning : Cytokeratins, biochemically related to intermediate filaments (IF), form an intracellular network of filaments which contributes to the mechanical stabilizing of the cell. 19 individual polypeptides, divided into two groups, constitute the cytokeratin family. LÄS MER
