  1. 1. Structure-function analysis of HAMLET (human alpha-lactalbumin made lethal to tumor cells)

    Författare :Jenny Pettersson; immunologi och glykobiologi - MIG Avdelningen för mikrobiologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; alpha-lactalbumin HAMLET oleic acid protein folding tumor cell death;

    Sammanfattning : The human genome sequence encodes fewer proteins than expected, suggesting that one protein can have several functions and adjust their structure to meet different structural demands. Changes in tertiary structure have mostly been associated with disease and the most striking example is the prion protein, which changes from a mixed alpha-helical and beta-sheet conformation to the beta-sheet rich, disease causing iso-form. 2. 2. Protein Complexity via Non-Native States: Binding, Stability, and Structural Studies of Calbindin D9k and HAMLET (Human alpha-lactalbumin Made LEthal to Tumor cells)

    Författare :Jonas Fast; Biofysikalisk kemi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; apoptosis; oleic acid; HAMLET; kinetic trap alpha-lactalbumin; calbindin; calcium; Metabolism; Biokemi; metabolism;

    Sammanfattning : Using optical spectroscopy, nuclear magnetic resonance (NMR), and differential scanning Calorimetry (DSC), I have studied two different calcium binding proteins that can form kinetically trapped altered states. Calcium is very important in numerous biological processes such as blood coagulation, signal transduction, muscle contraction and bone formation. LÄS MER

  3. 3. Apoptosis and tumor cell death pathways in response to HAMLET

    Författare :Oskar Hallgren; allergologi och palliativ medicin Lungmedicin; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biomedicinska vetenskaper allmänt ; General biomedical sciences; Klinisk biologi; apoptosis; autophagy; alpha-lactalbumin; Clinical biology; programmed cell death; HAMLET; cell cycle;

    Sammanfattning : HAMLET (Human alpha-lactalbumin made lethal to tumor cells) is a molecular complex of alpha-lactalbumin and oleic acid, that induces programmed cells death in tumor cells but leaves healthy differentiated cells unaffected. It has a broad anti-tumor activity as more than 40 cell lines from different origin are sensitive. LÄS MER

  4. 4. Cellular targets of HAMLET, their role in tumor cell death and therapeutic potential

    Författare :Aftab Nadeem; immunologi och glykobiologi - MIG Avdelningen för mikrobiologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cancer therapy; colon cancer; bladder cancer; cell death; HAMLET; alpha-lactalbumin; oleate; membrane lipids; nucleotide-binding protein; Ras GTPases; alpha-helical peptides;

    Sammanfattning : Protein-lipid complexes have broad and specific effects against cancers of different origins. HAMLET (Human Alpha-lactalbumin Made LEthal to Tumor cells) is a complex of partially unfolded α-lactalbumin and oleic acid that kills a wide range of tumor cells but leaves healthy differentiated cells unaffected. 5. 5. HAMLET - In vivo effects and mechanisms of tumor-cell death

    Författare :Lotta Gustafsson; immunologi och glykobiologi - MIG Avdelningen för mikrobiologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biomedicinska vetenskaper; Biomedical sciences; proteasome stress; HAMLET; human milk; tumor-cell death; clinical trial; rat xenograft model; skin papilloma; glioblastoma;

    Sammanfattning : HAMLET (human alpha-lactalbumin made lethal to tumor cells), a molecular complex derived from human milk, is an interesting new tool in cancer research since it induces programmed cell death in tumor cells while leaving normal, differentiated cells unharmed. The in vivo effects of HAMLET were studied in a rat xenograft model of human glioblastoma. LÄS MER
