  1. 21. Approaches to Optimizing High Content Confocal Microscopy

    Författare :Erland Lewin; Hjalmar Brismar; Kjell S. Carlsson; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES;

    Sammanfattning : This thesis presents two methods for improving high contentconfocal microscopy.The author's part in the first, "Toward a confocal subcellular atlasof the human proteome" was automating image capture of foursimultaneously tagged structures in cells in 96 well plates. In total,thousands of images of hundreds of proteins in cells. LÄS MER

  2. 22. Databases for antibody-based proteomics

    Författare :Erik Björling; Mathias Uhlén; Alvis Brazma; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; proteomics; antibodies; database; biomarker; website; Bioengineering; Bioteknik;

    Sammanfattning : Humans are believed to have ~20,500 protein-coding genes andmuch effort has over the last years been put into the characterizationand localization of the encoded proteins in order to understand theirfunctions. One such effort is the Human Proteome Resource (HPR)project, started in Sweden 2003 with the aim to generate specificantibodies to each human protein and to use those antibodies toanalyze the human proteome by screening human tissues and cells. Författare :Cecilia Lindskog Bergström; Fredrik Pontén; Anna Asplund; Ari Ristimäki; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Tissue microarrays; Antibody-based proteomics; Immunohistochemistry; Biomarker discovery; Diabetes; Lung cancer; Influenza virus; Evolution; Pathology; Patologi;

    Sammanfattning : Proteins are essential building blocks in every living cell, and since the complete human genome was sequenced in 2004, researchers have attempted to map the human proteome, which is the functional representation of the genome. One such initiative is the Human Protein Atlas programme (HPA), which generates monospecific antibodies towards all human proteins and uses these for high-throughput tissue profiling on tissue microarrays (TMAs). 4. 24. Finding order in chaos : Dissecting single-cell heterogeneity in space and time

    Författare :Christian Gnann; Emma Lundberg; Manuel D Leonetti; Lucas Pelkmans; KTH; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; non-genetic single-cell heterogeneity; cell cycle; metabolism; imaging-based subcellular proteomics; phosphoproteomics; icke-genetisk single-cell heterogenitet; cellcykel; metabolism; bildbaserad subcellulär proteomik; fosfoproteomik; Biotechnology; Bioteknologi;

    Sammanfattning : The cell is the smallest unit of life and contains DNA, RNA, proteins and a variety of other macromolecules. In recent years, technological advances in the field of single cell biology have revealed a staggering amount of phenotypic heterogeneity between cells in a population, which were previously considered homogenous. 5. 25. Exploring Biological Systems using Spatial Transcriptomic Technologies

    Författare :Kim Thrane; Joakim Lundeberg; Patrik Ståhl; Ulf Gyllensten; KTH; []
    Nyckelord :Spatial Transcriptomics; single-cell RNA-seq; scRNA-seq; spatial VDJ; Spatial Isoform Transcriptomics; Biotechnology; Bioteknologi;

    Sammanfattning : The transcriptome and the cells’ spatial organization are important determinants for the functions of biological systems, such as a tumor, brain, or skin tissue. Single-cell RNA sequencing (scRNA-seq) has emerged as a powerful tool for profiling the transcriptome of individual cells. LÄS MER
