1. DNA-Complexes with Drugs and Proteins
Sammanfattning : DNA is fundamental for all living cells; the DNA holds the genetic code, which is more or less the instruction book for how all cells are built and function. Several diseases are also linked to DNA, emerging either from a mutation in our genome, which could result in a malfunctioning protein, or that the transcription of genes is somehow affected by structural changes in the DNA, induced by mutations or DNA binding molecules. 2. Structural Adaptation of DNA to Small Ligands and Recombination Proteins
Sammanfattning : In order to contribute to the understanding of mechanisms of recognition in nucleic acid systems, the interaction of DNA with a variety of ligands, representing different cases of adaptation of the double-helix structure, was studied using flow linear dichroism (LD), circular dichroism (CD), fluorescence and absorption spectroscopic techniques. A chiral hydrophobic metal complex, [Ru(dpphen)3]2+ was examined, since this had been reported to be the most efficient molecule to discriminate DNA handedness. LÄS MER
3. Expression and regulation of Rad51 in human cells
Sammanfattning : Double strand breaks (DSBs) can be caused by exogenous DNA damaging agents or by endogenous processes, and may lead to chromosomal breakage and rearrangement resulting in apoptosis or tumorigenesis. The repair of DSBs in higher eukaryotes is to a large extent accomplished by homologous recombination (HR), a process in which DNA from a complementary strand is used as a template for repair synthesis. 4. Analysis of global gene expression in complex biological systems using microarray technology
Sammanfattning : The general aim of this thesis has been to explore the use of Affymetrix microarray technology in studies of global gene expression in complex biological systems, particularly with regard to the dynamics and heterogeneity of gene expression. The effects and systems studied are: 1. LÄS MER
