   Sammanfattning : Osteoporosis is a systemic, skeletal disease with the consequence of an increased risk of sustaining a fracture, one of the most common fractures being a vertebral fracture.    Sammanfattning : Studies on the epidemiology of distal femur fractures show a vast increase in low-energy trauma-associated fractures in older ages. Age-related changes in bones lead to loss of bone matrix and increased porosity, which entails an increased risk for fractures and different fracture patterns than in younger individuals. LÄS MER

   Sammanfattning : The purpose of this study was to (I) assess the association between body mass index (BMI) and total hip replacement (THR) and total knee replacement (TKR) due to osteoarthritis (OA), (II) assess the association between the mechanical load of work and THR and TKR due to OA, (III) evaluate the natural history of radiographic hip OA with regards to THR and hip fracture, (IV) to determine the prevalence of radiographic OA in patients with hip fracture, and (V) to examine and compare the inheritance patterns of THR for OA and hip fracture.    Sammanfattning : In this thesis, I evaluated the impact of pre-centrifugation sample management on the plasma metabolome, providing verification for the protocol used in a study where I applied metabolomics to better understand the role of body composition in fracture occurrence using a large population-based cohort.  In paper 1, I applied untargeted liquid chromatography-mass spectrometry (LC-MS) metabolomics analysis to plasma samples (n = 471) from healthy donors to predict and evaluate the effect of pre-centrifugation temperature and delay time on metabolomics data using a combination of random forest and generalized linear models. Hip fracture is one of the diagnoses that increases with age and it has become a major problem, both for those suffering a fracture and for society due to the large numbers involved, the morbidity with complications such as falls, functional decline, and the high mortality rate among those affected.
