  1. 1. The Hepatobiliary Transport of Rosuvastatin In Vivo

    Författare :Ebba Bergman; Hans Lennernäs; Ulf Bondesson; Jack Cook; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Rosuvastatin; Biliary excretion; Transport inhibition; Pharmacokinetics; Drug-drug interactions; Hepatobiliary transport; Organic anion transporting polypeptide; OATP; Statins; Gemfibrozil; Cyclosporine; Imatinib; Rifampicin; Canalicular transport; Sinusoidal transport; Hepatic uptake; Hepatic extraction; Biopharmacy; Biofarmaci; Biopharmaceutics; biofarmaci;

    Sammanfattning : In vivo studies of hepatobiliary disposition are challenging. The hepatobiliary system is complex, as its physiological localization, complex cellular structure with numerous transporters and enzymes, and the interindividual variability in protein expression and biliary flow will all affect the in vivo disposition of a drug under investigation. 2. 2. In vitro and in silico Predictions of Hepatic Transporter-Mediated Drug Clearance and Drug-Drug Interactions in vivo

    Författare :Anna Vildhede; Per Artursson; Maria Karlgren; Maria Kjellsson; Ulf Norinder; Kim Brouwer; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; OATP1B1; OATP1B3; OATP2B1; NTCP; drug transporters; human hepatocytes; atorvastatin; pitavastatin; proteomics; sandwich-cultured human hepatocytes; SCHH; mechanistic modeling; in vitro-in vivo extrapolation; transport inhibition; hepatic uptake; hepatocyte isolation; transporter contribution; Biopharmaceutics; Biofarmaci;

    Sammanfattning : The liver is the major detoxifying organ, clearing the blood from drugs and other xenobiotics. The extent of hepatic clearance (CL) determines drug exposure and hence, the efficacy and toxicity associated with exposure. Drug-drug interactions (DDIs) that alter the hepatic CL may cause more or less severe outcomes, such as adverse drug reactions. LÄS MER

  3. 3. Hepatic and Peritoneal Colorectal Metastases : Aspects of Prognosis and Treatment

    Författare :Haile Mahteme; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Surgery; Colorectal cancer; lever metastas; peritoneal metastas; intraperitoneal; tumör reduktion; Kirurgi; Surgery; Kirurgi; Surgery; kirurgi;

    Sammanfattning : Although two-thirds of colorectal cancer patients are cured by surgery, approximately 50% of the patients with this disease develop locally recurrent or distant metastases during the course of their illness. The aim of this study was to identify metastatic sites associated with poor prognosis in rectal cancer and then to investigate methods that can prevent the development and growth of metastases and optimise uptake of drugs at these sites in animal models. 4. 4. Adipose tissue inflammation, hepatic fat accumulation and insulin resistance

    Författare :Maria Kolak; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Insulin resistance is the central feature of a variety of metabolic disorders, such as type 2 diabetes and the metabolic syndrome. The incidence of these disorders, often together with obesity, is currently increasing throughout the world. LÄS MER

  5. 5. Studies on the regulation of hepatic and intestinal lipid metabolism

    Författare :Kristina Kannisto; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Disturbances in the lipid homeostasis can result in a wide variety of different diseases and symptoms. This thesis is focused on the regulation of cholesterol metabolism and its relation to atherosclerosis development, and the effect of mitochondrial (mt)DNA depletion on hepatic β-oxidation of fatty acids.
