  1. 1. Theoretical Studies of Cytochrome P450

    Författare :Patrik Rydberg; Beräkningskemi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; quantum chemistry; Proteins; enzymology; Proteiner; enzymologi; Theoretical chemistry; transition-state force field; peroxidase; theoretical chemistry; molecular dynamics; density functional theory; cytochrome P450; porphyrin; haem; heme; kvantkemi; Teoretisk kemi;

    Sammanfattning : The cytochromes P450 are a large enzyme family that is found in all living organisms and takes part in both endogenous and exogenous metabolism. They are primarily mono-oxygenases and perform a wide range of reactions. LÄS MER

  2. 2. Theoretical studies of porphyrin proteins

    Författare :Emma Sigfridsson; Beräkningskemi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; molecular mechanics; quantum chemical calculations; electrostatic potential charges; atomic charges; ferrochelatase; cytochrome; myoglobin; Porphyrin; haem; QM MM; reorganisation energy; Theoretical chemistry; quantum chemistry; Teoretisk kemi; kvantkemi;

    Sammanfattning : Different aspects of porphyrin proteins have been studied with theoretical methods. For example, we have studied: - The importance of hydrogen bonds for the discrimination between carbon monoxide and molecular oxygen by myoglobin, the oxygen carrier in muscles, modelling the active site both in vacuum and in the protein matrix - The inner-sphere reorganisation energy during electron transfer for cytochromes compared to other electron-transfer proteins, such as blue copper proteins and Fe-S clusters - The role of porphyrin distortions in the reaction mechanism of ferrochelatase, the enzyme that inserts an iron ion into protoporphyrin IX to make haem - The interaction between different metal sites in ferrochelatase In these studies we have used density functional theory, classical force field methods, and a combination of quantum chemistry and molecular mechanics. 3. 3. Exploring Brain Gene Expression i Animal Models of Behaviour

    Författare :Julia Lindberg; Elena Jazin; Peter Saetre; Jonathan Flint; Uppsala universitet; []
    Nyckelord :Biology; behavioural genetics; gene expression; brain; microarray analysis; domestication; animal model; haem; Canis familiaris; Vulpes vulpes; Mus musculus; Biologi;

    Sammanfattning : The genetic basis for behavioural traits is largely unknown. The overall aim of this thesis was to find genes with importance for behavioural traits related to fear and anxiety. Microarray analysis was used to screen expression profiles of brain regions important for emotional behaviour in dogs, wolves, foxes and mice. LÄS MER

  4. 4. Iron in Haem Biosynthesis. Structural Studies of Ferrochelatase and Frataxin

    Författare :Tobias Karlberg; Biokemi och Strukturbiologi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Naturvetenskap; Natural science; Molekylär biofysik; Molecular biophysics; Biomedicinska vetenskaper; Biomedical sciences; protein crystallography; ferrochelatase; haem biosynthesis; iron; frataxin;

    Sammanfattning : Ferrochelatase catalyses the synthesis of heme by inserting Fe(II) into protoporphyrin IX. The structure of ferrochelatase from yeast has been determined (2.4 Å). The structure is homodimeric with each of the monomer made up of two Rossmann type domains with an active-site cleft between them. LÄS MER

  5. 5. Transmembrane proton transfer in biological systems : an investigation of bacterial and archaeal haem-copper oxidases

    Författare :Gwen Gilderson; Ólöf Einarsdóttir; Stockholms universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biochemistry; Biokemi; biokemi; Biochemistry;

    Sammanfattning : Haem-copper oxidases are membrane-bound protein complexes that catalyse the reduction of dioxygen to water in the respiratory chain. These enzyme complexes are molecular machines that couple the free energy released from this reaction to pump protons across the membrane. LÄS MER
