  1. 1. Studies on mutagenesis by ethylene oxide and styrene oxide in the hprt gene in human cells

    Författare :Tatiana Bastlova; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :ethylene oxide; styrene; styrene-7; 8-oxide; human diploid fibroblasts; human T-lymphocytes; hprt mutation;

    Sammanfattning : Environmental, occupational and life style-related exposures to mutagenic agents may contribute to cancer risk in humans. To prevent the potentially hazardous effects of such agents it is important to understand their mechanisms of action. LÄS MER

  2. 2. Mechanistic studies on forward and reverse mutations in the hprt gene of Chinese hamster cells

    Författare :Li-Hua Zhang; Stockholms universitet; []
    Nyckelord :;

  3. 3. HPRT mutational spectra and microsatellite DNA instability in HNPCC and lung cancer patients

    Författare :Peter Hackman; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Somatic gene mutation; mutational spectra; HPRT gene; lung cancer; smoking; hMLH1 gene; HNPCC; microsatellite instability; mismatch repair; T-cells;

    Sammanfattning : The, general aims of this work have been to explore the use of microsatellite DNA length variation and mutational spectra of the hypoxanthine guanine phosphoribosyltransferase (HPRT) locus in T-cells as tools for a better understanding of human somatic mutagenesis in vivo. In particular (a) how inherited mismatch repair (MMR) deficiency may affect the stability of microsatellite DNA and the mutational spectrum at the HPRT locus, and (b) how the mutational spectrum at the HPRT locus is influenced by smoking, and by predisposition to lung cancer among never smokers. 4. 4. Environmental Contaminants, Recombination and Cancer : Investigations on the molecular mechanisms of spontaneous and induced recombination in the hprt gene of Chinese hamster cells

    Författare :Thomas Helleday; Mark Meuth; Stockholms universitet; []
    Nyckelord :toxikologisk genetik; Genetic Toxicology;

    Sammanfattning : Recombination results in rearrangement of the genetic material contained in the chromosomes inherited from the mother and father and this process is also involved in repairing damage to the genome. Occasionally, the machinery that performs recombination malfunctions and this may lead to illegitimate recombination events which inactivate tumor suppressor genes or activate protooncogenes. 5. 5. Mutation profile at the hprt locus in T-cells of non-smoking males

    Författare :Anne-May Österholm; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :human T-cells; hprt mutation; in vivo; deletion; splicing; base substitution;

    Sammanfattning : Extensive studies have been done on mutations in several marker genes in order to predict health consequences such as carcinogenesis. However, more information about mutations arising in vivo in somatic cells is needed to understand the relationship between the different factors involved in mutagenesis such as DNA damage, DNA repair capacity and individual susceptibility, and to elucidate the influence of endogenous mechanisms and environmental exposures.
