11. Clinical Pharmacokinetics of the Antimalarial Artemisinin Based on Saliva Sampling
Sammanfattning : Artemisinin is the parent compound of a novel family of antimalarials. Repetitive administrations of artemisinin to both healthy volunteers and malaria patients have been shown to result in decreased plasma concentrations of the compound, most probably due to an autoinduction of different CYP450 enzymes. 12. Nucleic Acids as Drug Targets -Interactions of Platinum Complexes, Polyamines, and siRNA with DNA and RNA Models
Sammanfattning : Drugs targeting nucleic acids were studied in DNA and RNA model systems. The effects were investigated with respect to structural and thermodynamic influence, kinetics, and biological function. The drugs employed in this thesis were anticancer active platinum(II) complexes, polyamine derivatives, and siRNA. LÄS MER
13. Pharmacokinetics of the enantiomers of thalidomide
Sammanfattning : Thalidomide has a chiral center and the racemate of (+)-(R) and (-)-(S)-thalidomide was introduced as a hypnotic/sedative drug in 1957. In 1961 it was withdrawn due to teratogenicity and neuropathy. There is a growing interest in thalidomide treatment of immunomodulatory diseases. LÄS MER
14. N-acetylaspartate in brain - studies on efflux and function
Sammanfattning : N-acetylaspartate (NAA) is an amino acid derivative present in high concentration in the brain. The function of NAA is still unsettled in spite of 50 years of research. The mainly neuronal synthesis and glial breakdown of NAA requires a well regulated neuronal efflux and glial uptake. LÄS MER
15. Interactions of Anticancer Active Platinum(II) Complexes with DNA
Sammanfattning : The kinetics and mechanism for interactions of the anticancer active platinum(II) complexes cis-[PtCl2(NH3)2)], cis-[PtCl2(NH3)(c-C6H11NH2)], and trans-[PtCl2-(NH3)(quinoline)] with DNA have been studied using a combination of HPLC, circular dichroism (CD), UV/vis spectroscopy, and differential scanning calorimetry (DSC). Reactions of the monoaqua forms (charge = +1) of these complexes with single-stranded DNA oligonucleotides, typically between 13-17 bases long, were found to depend on i) the composition of the reaction medium, ii) the exact nature and location of the platination target, and iii) on the DNA structure.
